Abstract The mechanisms of interaction and crosstalk between tumor cells tumor-associated macrophages (TAMs) have provided novel options for intervening in progression. However, the molecular TAMs underlying oral squamous cell carcinoma (OSCC) invasio, migration chemoresistance remain unclear. This study sought to specifically investigate role tumor-cell-derived paracrine heat shock protein 27 (HSP27) OSCC invasion, potential TAMs. In this study, bioinformatic analysis IHC results demonstrated that expression level HSP27 was higher tissues patients with advanced lymph node metastasis than early stage non-metastatic patients, its positively correlated levels multidrug resistance-associated proteins macrophage infiltration. vivo, Survival low-expressing xenograft model mice inferior controls. vitro, TAMs-CM significantly up-regulated two types including CAL27 SCC9 cells. cell-derived regulated through mode reduced apoptosis induced by chemotherapeutic drug cisplatin cells, thus promoting OSCC. promoted secretion cytokine IL-6 from TAMs, whereas TAMs-derived enhanced chemoresistance, invasion an autocrine activates β-catenin pathway during process, additionally stem properties manner. Abstract Hypoxia-inducible factors (HIFs) are essential transcription that orchestrate cellular responses to oxygen deprivation. HIF-1α, as an unstable subunit of HIF-1, is usually hydroxylated by prolyl hydroxylase domain enzymes under normoxic conditions, leading ubiquitination and proteasomal degradation, thereby keeping low levels. Instead hypoxia, sometimes even in normoxia, HIF-1α translocates into the nucleus, dimerizes with HIF-1β generate then activates genes involved adaptive such angiogenesis, metabolic reprogramming, survival, which presents new challenges insights its role processes. Thus, review delves mechanisms HIF-1 maintains stability normoxia including but not limited giving transcriptional, translational, well posttranslational regulation underscore pivotal adaptation malignancy. Moreover, extensively cancer cardiovascular diseases potentially serves a bridge between them. An overview HIF-1-related drugs approved or clinical trials summarized, highlighting their potential capacity for targeting toxicity related treatment. Breast cancer (BC) is characterized by an extensive genotypic and phenotypic heterogeneity. In-depth investigations into the molecular bases of BC phenotypes, carcinogenesis, progression, metastasis are necessary for accurate diagnoses, prognoses, therapy assessments in predictive, precision, personalized oncology. This review discusses both classic as well several novel omics fields that involved or should be used modern investigations, which may integrated a holistic term, onco-breastomics. Rapid recent advances profiling strategies analytical techniques based on high-throughput sequencing mass spectrometry (MS) development have generated large-scale multi-omics datasets, mainly emerging from three ”big omics”, central dogma biology: genomics, transcriptomics, proteomics. Metabolomics-based approaches also reflect dynamic response cells to genetic modifications. Interactomics promotes view research constructing characterizing protein–protein interaction (PPI) networks provide hypothesis pathophysiological processes progression subtyping. The emergence new omics- epiomics-based multidimensional opportunities gain insights heterogeneity its underlying mechanisms. main epiomics (epigenomics, epitranscriptomics, epiproteomics) focused epigenetic DNA changes, RNAs modifications, posttranslational modifications (PTMs) affecting protein functions in-depth understanding cell proliferation, migration, invasion. Novel fields, such epichaperomics epimetabolomics, could investigate interactome induced stressors PPI metabolites, drivers BC-causing phenotypes. Over last years, proteomics-derived omics, matrisomics, exosomics, secretomics, kinomics, phosphoproteomics, immunomics, provided valuable data deep dysregulated pathways their tumor microenvironment (TME) immune (TIMW). The review describes correlations between impaired functioning of chaperones and co-chaperones in Alzheimer's disease (AD) pathogenesis. study aims to highlight significant lines research this field. Chaperones like Hsp90 or Hsp70 are critical agents regulating cell homeostasis. Due some conditions, aging, their activity is damaged, resulting β-amyloid tau aggregation. This leads the development neurocognitive impairment. Dysregulation one causes condition. Disorders molecules PP5, Cdc37, CacyBP/SIPTRAP1, CHIP protein, FKBP52, STIP1 play a key role AD FKBP52 co-chaperones. protein co-chaperone that switches Hsp70/Hsp90 complexes, binds Hsp70. Recognition precise processes allows for invention effective treatment methods. Potential drugs may either reduce levels inhibit accumulation Some substances neuroprotect from Aβ toxicity. Acid tolerance is achieved through internal pH regulation, acid efflux pumps, proteins, allowing in acidic matrices the gastrointestinal tract.
