Clinical studies have shown that epileptic seizures worsen Alzheimer's disease (AD) pathology and related cognitive deficits; however, the underlying mechanism is unclear. To assess effects of on progression AD, chronic temporal lobe epilepsy was induced in five familial AD mutation (5×FAD) mice by kindling with chemoconvulsant pentylenetetrazole (PTZ) at 3-3.5 months age. The amyloidogenic pathway, tauopathy, synaptic damage, neuronal death, neurological inflammatory response associated kinase signaling pathway dysregulation were examined 9 We found APP, p-APP, BACE1, Aβ kinase-associated p-tau levels elevated after PTZ 5×FAD mice. In addition, exacerbated hippocampal damage cell as determined scanning electron microscopy terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling (TUNEL) staining, respectively. Finally, neuroinflammation markers GFAP Iba1, well cytokine IL-1β, increased insult. profoundly extracellular regulated (ERK)-death-associated protein (DAPK) overactivation, acute ERK inhibitor treatment downregulated production p-APP long-term use antiseizure drug carbamazepine (CBZ) alleviated seizure-induced accelerated amyloid tau ERK-DAPK overactivation Collectively, these results demonstrate increases AD-like neuropathology are partially suggesting axis could be a new therapeutic target for patients comorbid seizures.

Background: Alzheimer’s disease (AD) is a type of frequently occurs in the elderly population. Diagnosis and treatment methods for this are still lacking, more research required. In addition, little known about function circular RNAs (circRNAs) AD. Methods: research, RNA expression data AD from Gene Expression Omnibus (GEO) database were downloaded. The levels circRNAs cerebrospinal fluid samples healthy participants patients measured by reverse transcription‑quantitative PCR (RT-qPCR). diagnosed value differential expressed was analyzed with Receiver operating characteristic curve (ROC). Pathways related to circ_0001535 found using gene set enrichment analysis (GSEA) Metascape. direct interactions between E2F transcription factor 1 (E2F1) or E2F1 dihydrofolate reductase (DHFR) verified Chromatin immunoprecipitation (ChIP) Binding Protein Immunoprecipitation (RIP) assays. Cell Counting Kit-8 (CCK8) flow cytometry used identify circ_0001535/E2F1/DHFR axis on proliferation apoptosis cells. Results: total, 12 have been linked diagnosis. 7 differed control groups. Circ_0001535 had most diagnose among these circRNAs. Hence, regarded as key circRNA present study. E2F1/DHFR predicted be regulated circ_0001535. IP assays experiment results showed that could bind promoter region DHFR vitro overexpression promote expression, while knock down inhibit SH-SY5Y Finally, rescue experiments suggested reduce Aβ25-35-induced cell facilitate through axis.
