Liver cancer represents the fourth leading cause of cancer-associated death worldwide. The heterogeneity its tumor microenvironment (TME) is a major contributing factor metastasis, relapse, and drug resistance. Regrettably, late diagnosis makes most liver patients ineligible for surgery, frequent failure non-surgical therapeutic options orientates clinical research to investigation new drugs. In this context, cellular senescence has been recently shown play pivotal role in progression chronic inflammatory diseases, ultimately cancer. Moreover, stem-like state triggered by associated with emergence drug-resistant, aggressive clones. recent years, an increasing number studies have emerged investigate senescence-associated hepatocarcinogenesis derived therapies, promising results. review, we intend provide overview evidence that unveils forms primary metastatic cancer, focusing on involvement mechanism therapy

Blunting the tumor's stress-sensing ability is an effective strategy for controlling tumor adaptive survival and metastasis. Here, we have designed a cyclically amplified nano-energy interference device based on lipid nanoparticles (LNP), focused altering cellular energy metabolism. This innovative nano efficiently targets monitors status while simultaneously inhibiting mitochondrial respiration, biogenesis ribosome production. To this end, first identified azelaic acid (AA), binary capable of disrupting respiratory chain. Upon encapsulation in LNP linkage to mitochondrial-targeting molecules, disruptive effect further augmented. Consequently, tumors exhibit substantial upregulation glycolytic pathway, intensifying their glucose demand worsening energy-deprived microenvironment. Then, analog, 2-Deoxy-D-glucose (2-DG), linked LNP, competitively inhibits normal uptake. The synergetic results combining AA with 2-DG induce comprehensive deficiency within tumors, blocking generation energy-sensitive ribosomes. Ultimately, disruption both mitochondria ribosomes depletes supply new protein-generating capacity, rendering adapt environmental stress thereby growth Comprehensively, device, by ability, provides novel therapeutic refractory tumors.

Colorectal cancer (CRC) exhibits a substantial morbidity and mortality rate, with its aetiology pathogenesis remain elusive. It holds significant importance within the tumour microenvironment (TME) exerts crucial regulatory influence on tumorigenesis, progression, metastasis. TAMs possess capability to foster CRC pathogenesis, proliferation, invasion, metastasis, as well angiogenesis, immune evasion, resistance. Furthermore, can mediate prognosis of CRC. In this paper, we review mechanisms by which natural compounds target exert anti-CRC effects from perspective promotional CRC, mainly regulating polarization TAMs, reducing infiltration recruitment enhancing phagocytosis macrophages, signalling pathways cytokines, discuss potential value therapeutic strategies compounds-targeting pathway in clinical treatment.

Colorectal malignancy is the third most common cancer and one of prevalent causes death globally. Around 20-25% patients present with metastases at time diagnosis, 50-60% develop in due course disease. Liver, followed by lung lymph nodes, are sites colorectal metastases. In such patients, 5-year survival rate approximately 19.2%. Although surgical resection primary mode managing metastases, only 10-25% competent for curative therapy. Hepatic insufficiency may be aftermath extensive hepatectomy. Hence formal assessment future liver remnant volume (FLR) imperative prior to surgery prevent hepatic failure. The evolution minimally invasive interventional radiological techniques has enhanced treatment algorithm Studies have demonstrated that these address limitations resection, as insufficient FLR, bi-lobar disease, higher risk surgery. This review focuses on palliative role through procedures including portal vein embolization, radioembolization, ablation. Colorectal cancer (CRC) commonly metastasizes to the liver, and this poses a significant clinical challenge. Tumor-associated macrophages (TAMs), key players within TME, play role in promoting CRC metastasis by secreting various chemokines, growth factors, cytokines. This review not only aims enhance our knowledge of TAMs' functions progression but also examines innovative therapeutic strategies address problem colorectal liver (CLM). By targeting TAMs, we may be able develop more effective treatments offer hope patients suffering from devastating disease.
