Abstract Dry eye disease (DED) affects up to half of the world’s population and causes blur, pain, decreased quality life. Unfortunately, many people do not receive proper treatment due current challenges associated with DED diagnoses. To address this issue, point-of-care (PoC) clinical tear film diagnostics are beginning be used for rapid in-office testing. However, these also have significant obstacles, including sampling. We previously demonstrated how soft contact lenses (SCLs) can capture concentrate proteins on eye. Given low volume subjects, SCL sampling holds great potential improve PoC facilitate prompt patients. Here we show detection new established protein biomarkers using our method. Our analysis revealed associations between immune system, ferroptosis, salivary secretion, calcium binding proteins. identified correlations testing proteomics, emphasizing power importance translational medicine research. These experiments lay foundation both sampled biomarker discovery as well improved diagnostic patient care. Figure

Dry eye disease (DED) is a multifactorial condition affecting the ocular surface. It characterized by loss of tear film homeostasis and accompanied symptoms that may potentially result in damage to surface even vision loss. Unmodifiable risk factors for DED mainly include aging, hormonal changes, lifestyle issues such as reduced sleep duration, increased screen exposure, smoking, ethanol consumption. As its prevalence continues rise, has garnered considerable attention, prompting exploration potential new therapeutic targets. Recent studies have found when production ROS exceeds capacity antioxidant defense system on surface, oxidative stress ensues, leading cellular apoptosis further damage. These events can exacerbate inflammation responses, increasing levels promoting vicious cycle DED. Therefore, given central role reactive oxygen species DED, strategies involving antioxidants emerged novel approach treatment. This review aims enhance our understanding intricate relationship between thereby providing directions explore innovative approaches this complex disorder.

Abstract Dry eye disease (DED) is a widespread, multifactorial, and chronic disorder of the ocular surface with disruption tear film homeostasis as its core trait. Conjunctival goblet cells (CGCs) are specialised secretory found in conjunctival epithelium that participate formation by secreting mucin. Changes both structure function CGCs hallmarks DED, imaging assessment important for diagnosis, classification, severity evaluation DED. Existing methods include biopsy, impression cytology vivo confocal microscopy, which can be used to assess morphology, distribution, density CGCs. Recently, moxifloxacin‐based fluorescence microscopy has emerged novel technique enables efficient, non‐invasive This article presents comprehensive overview their alterations context well current assessment. Ferroptosis plays an important role in the occurrence and development of many diseases, including neurodegenerative diseases. Thus, ferroptosis inhibitors able to cross blood-brain barrier may have therapeutic potential. The best so far are lipophilic radical trapping antioxidants (RTAs) that block lipid peroxidation membranes. Several generations ferrostatins been synthesized, among which UAMC-3203 showed high potency animal models with improved properties compared ferrostatin-1. To further improve its pharmacokinetics properties, drug-likeness, permeability, we modified by decreasing size molecule reducing polarity replacing sulfonamide first amide groups subsequently isosteric oxazoles. Herein, present design, synthesis, biological evaluation a novel series oxazole RTAs potency, excellent oral bioavailability, concentrations brain tissue.

Dry eye disease (DED) is a chronically inflammatory ocular surface disorder of unknown pathogenesis. Anti-inflammatory medications, artificial tears, autologous serum, and LipiFlow have been shown to be highly beneficial in alleviating symptoms. Nevertheless, these interventions often provide only short-term results do not address the underlying problems disease. There growing evidence that risk DED associated with vicious cycle inflammation. This inflammation produced by interaction several factors, including tear film hyperosmolarity, instability, inflammation, apoptosis. Chinese medicine monomers, distinguished their multicomponent multitarget advantages, help treat modulating status, inhibiting responses, apoptosis, providing new way thinking management medicine.

Dry eye disease (DED) is a chronic multifactorial ocular surface mainly caused by the instability of tear film, characterized series discomforts and even visual disorders. Oxidative stress has been recognized as an upstream factor in DED development. Diquafosol sodium (DQS) agonist P2Y2 receptor to restore integrity/stability film. With ability alternate between Ce3+ Ce4+, cerium oxide nanozymes could scavenge overexpressed reactive oxygen species (ROS). Hence, DQS-loaded nanozyme was designed boost synergistic treatment DED. Cerium with branched polyethylenimine-graft-poly(ethylene glycol) nucleating agent dispersant fabricated followed DQS immobilization via dynamic phenylborate ester bond, obtaining (defined Ce@PBD). Because mimic cascade processes superoxide dismutase catalase, Ce@PBD excessive accumulated ROS, showing strong antioxidant anti-inflammatory properties. Meanwhile, receptors conjunctival cells be stimulated Ce@PBD, which can relieve incompleteness The animal experiments demonstrated that significantly restored defect corneal epithelium increased number goblet cells, promotion secretion, best among commercial ophthalmic solutions.
