Neurodegenerative diseases are a large class of neurological disorders characterized by progressive dysfunction and death neurones. Examples include Alzheimer's disease, Parkinson's frontotemporal dementia, amyotrophic lateral sclerosis. Aging is the primary risk factor for neurodegeneration; individuals over 65 more likely to suffer from neurodegenerative with prevalence increasing age. As population ages, social economic burden caused these will increase. Therefore, new therapies that address both aging neurodegeneration imperative. Ketogenic diets (KDs) low carbohydrate, high-fat developed initially as an alternative treatment epilepsy. The classic ketogenic diet provides energy via long-chain fatty acids (LCFAs); naturally occurring medium chain (MCFAs), on other hand, main components medium-chain triglyceride (MCT) diet. MCT-based efficient at generating ketone bodies used secondary source neurones astrocytes. However, levels alone do not closely correlate improved clinical symptoms. Recent findings suggest mode action MCFAs, e.g., improving mitochondrial biogenesis glutamate receptor inhibition. MCFAs have been linked disease their effects metabolism. Oxidative stress is involved in a wide range of age-related diseases. A critical role has been proposed for mitochondrial oxidative initiating or promoting these pathologies and the potential mitochondria-targeted antioxidants to fight them, making their search testing very urgent task. In this study, SkQ1, SkQ3 MitoQ were examined as they affected isolated rat liver mitochondria yeast cells, comparing with clinically tested SkQ1 MitoQ. At low concentrations, all three substances stimulated oxidation respiratory substrates state 4 respiration (no ADP addition); at higher inhibited ADP-triggered 3 uncoupled state, depolarized inner membrane, contributed opening mPTP (mitochondrial permeability transition pore), did not specifically affect ATP synthase, had pronounced antioxidant effect. was most active antioxidant, possessing, unlike MitoQ, prooxidant activity increasing concentrations. reduced prooxidant-induced intracellular cell death prevented reversed fragmentation, being efficient. These data allow us consider promising therapeutic agent mitigate associated stress.

Alzheimer’s disease (AD) is an age-related, multifaceted neurological disorder associated with accumulation of aggregated proteins (amyloid Aβ and hyperphosphorylated tau), loss synapses neurons, alterations in microglia. AD was recognized by the World Health Organization as a global public health priority. The pursuit better understanding forced researchers to pay attention well-defined single-celled yeasts. Yeasts, despite obvious limitations application neuroscience, show high preservation basic biological processes all eukaryotic organisms offer great advantages over other models due simplicity, growth rates on low-cost substrates, relatively simple genetic manipulations, large knowledge base data collections, availability unprecedented amount genomic proteomic toolboxes high-throughput screening techniques, inaccessible higher organisms. Research reviewed above clearly indicates that yeast models, together other, more including animal C. elegans Drosophila, significantly contributed tau biology. These allowed throughput factors drugs interfere oligomerization, aggregation toxicity, hyperphosphorylation. In future, will remain relevant, focus creating novel systems facilitate identification earliest biomarkers among different cellular networks order achieve main goal—to develop new promising therapeutic strategies treat or prevent disease.

Quinone derivatives of triphenylphosphonium have proven themselves to be effective geroprotectors and antioxidants that prevent the oxidation cell components with participation active free radicals – peroxide (RO2•), alkoxy (RO•), alkyl (R•), as well reactive oxygen species (superoxide anion, singlet oxygen). The most studied representatives are plastoquinone (SkQ1) ubiquinone (MitoQ), which in addition antioxidant properties also had a strong antibacterial effect. Other quinone based on decyltriphenylphosphonium (SkQ3, SkQT, SkQThy) been less their activity remains undiscovered. In this work, we investigated decyltriphenylphosphonium. We shown that, case SkQ1, they against various gram-positive bacteria, while not so gram-negative bacteria. This is associated recognition by main multidrug resistance pump bacteria AcrAB-TolC. Moreover, SkQ1 itself, there dependence its action number bacterial cells molar amount antibiotic distinguishes it from conventional antibiotics. At same time, cytotoxic effect mammalian observed higher concentrations, suggests presence protective caused separation process energy generation transport membrane. Another possible explanation could different composition multidrug-resistant (MDR) pumps. difference effects may due expression profiles MDR pumps types.
