To preserve male fertility after diagnosis of any kind cancer, a prompt assessment the semen quality and an appropriate cryopreservation must be performed before radio-chemotherapy starts. The present work aims to evaluate parameters at different cancer patients thawing. Testicular tumors lymphomas are among most common cancers in younger patients, while chemotherapy significantly increases patients' survival, it can epigenetically alter fluid, resulting temporary or permanent infertility. We analyzed data from database Gamete Cryopreservation Center (Annunziata Hospital, CS; Italy) period 2011-2020 cohort 254 aged 18-56 years. evaluation was blind manner anonymously recovered; main referring were assessed accordance with WHO guidelines decision limits (6th edition; 2021). types as follows: testis (TC; n = 135; 53.1%), hematological (HC; 76; 29.9%), other (OC; 43; 17%). Comparing TC vs. HC (P1) OC (P2), had worst quality: sperm number/mL (P1 0.0014; P2 0.004), total motility 0.02; 0.07), progressive 0.04; 0.05), viability 0.01; 0.02), percentage atypical morphology 0.05; 0.03). After thawing, recovery lowered, accounting for 46.82% 16.75%, respectively, whole cohort; similarly, subgroups ascribed TC, they showed lowest recovery. Strong correlation existed between pre- post-cryopreservation (p < 0.001) subgroup 0.05). All subgroups, extents, findings below reference values, suggesting diverse susceptibilities cryodamage. Cancer associated treatments affect quality, meaning should considered useful personalized prerogative timely manner.

Semen cryopreservation has played an important role in medically assisted reproduction for decades. In addition to preserving male fertility, it is sometimes used overcoming logistical issues. Despite its proven clinical usability and safety, there a lack of knowledge how affects spermatozoa at the molecular level, especially terms non-coding RNAs. Therefore, we conducted this study, where compared slow freezing vitrification good- poor-quality human semen samples by analyzing conventional sperm quality parameters, performing functional tests expression miRNAs. The results revealed that normozoospermic does not alter maturity (protamine staining, hyaluronan binding), although can increase DNA fragmentation lower motility. On both types cryopreservation, miRNAs from are significantly overexpressed those native patients, but oligozoospermic samples, effect observed only after vitrification. Moreover, show selected mostly samples. Conversely, when vitrified were compared, determined miR-99b-5p was comparing freezing, miR-15b-5p miR-34b-3p under-expressed our imply modulate miRNA profiles become comparable

Sperm cryopreservation provides patients undergoing oncological, surgical, or infertility treatments the opportunity to conceive their own children, using assisted reproductive technologies. However, freezing-thawing process can negatively influence both quantity and quality of spermatozoa, mainly due an excessive production reactive oxygen species and/or impaired antioxidant defense system in sperm. Aromatic medicinal plants synthesize essential oils with proprieties as a part ecological adaptation environmental stress, thanks rich bioactive phytochemical components. This study aimed assess sperm progressive motility, viability, plasma membrane functionality, lipid peroxidation levels human cryopreserved normozoospermic (n = 51) asthenozoospermic samples without addition Thymus satureoides (TSEO) (20 µg/mL), Artemisia vulgaris (AVEO) (48 Rosmarinus officinalis (ROEO) (13 µg/mL) oils. parameters were significantly better preserved ROEO (p < 0.05) 0.01). In contrast, TSEO had negative impact for groups 0.05). Meanwhile, no significant effects observed AVEO. summary, revealed that vitro agents during be either beneficial, which helps preserve fertilizing potential, detrimental spermicidal agents.

Background/Objectives: Semen cryopreservation has been widely used in recent decades, mainly patients with male factor infertility (MFI) and cancer (to preserve their fertility potential before undergoing gonadotoxic treatments, such as chemotherapy or radiotherapy). Methods: we evaluated the temporal trend existence of factors determining usage cryopreserved semen (in order, for instance, to conceive using assisted reproduction techniques (ARTs)). We 7044 who since 1991, evaluating clinical information diagnosis, age, method which collection took place number pick-ups use ART. Results: The mean age was 33.12 ± 8.41 years. most common cancers were testicular (1758/3262) hematological (942/3262). Patients underwent MFI picked up samples much more than neoplastic (42.15% vs. 8.55%). 35-40 years old frequently when compared other groups. Moreover, found several data regarding sub-types methods collection. Conclusions: According our results, indication cryopreservation, might represent a simple way predict future

ABSTRACT The association between suboptimal sperm parameters and DNA damage in mature spermatozoa indicates that some individuals may have intrinsic issues with spermatogenesis. objective of this study is to evaluate the impact cryopreservation reactive oxygen species (ROS) on stability, as assessed by Sperm Chromatin Structure Assay (SCSA) motility viability, ICSI Result. Participants contacted Fertility Clinic provided written informed consent. All data were subsequently handled processed a confidential anonymous manner. There was source 300 participants (600 semen samples) study, divided into 3 groups. female partners underwent ICSI, (some oocytes injected fresh sperms others cryopreserved for same patient (liquid nitrogen at−196°C, accordance conventional protocols). Cryopreservation from fertile men does not seem adversely affect integrity either or prepared samples. However, morphology both samples significantly compromised process. No evidence cryopreserving healthy males would compromise genetic material laboratory-prepared

However, impact size effects on crystal formation, growth, performance has been rarely explored. Here, graphene oxide nanosheets (GO NSs) with controllable sizes were used as a representative 2D probe regulation rewarming in cell cryopreservation. All GO NSs exhibited notable inhibitory during recrystallization process. Significantly, when was smaller than certain (<150 nm), they showed more significant suppression effects, which could reduce about 17% that pure water. Meanwhile, experiments also indicated smaller-sized better behavior, 90 nm (GO-90) heating 70 °C just 1 min induced by an 808 laser (2 W/cm

For male patients, therapeutic option preserve fertility is semen cryopreservation. Among tests evaluating sDF, TUNEL (Terminal deoxynucleotidyl transferase dUTP nick end labeling) and SCD (Sperm Chromatin Dispersion) are widely used. Our lab developed a modified version of (TUNEL/PI) able distinguish two populations (PI Brighter PI Dimmer) differently associated with viability reproductive outcomes. The aim this study was compare sDF levels detected by TUNEL/PI the samples from 71 subjects attending our Andrology Laboratory. results demonstrate that is less sensitive determining compared TUNEL/PI. statistically significant positive correlation found between evaluated Dimmer (consisting all dead spermatozoa) suggests mainly detects unviable spermatozoa.
