To preserve male fertility after diagnosis of any kind cancer, a prompt assessment the semen quality and an appropriate cryopreservation must be performed before radio-chemotherapy starts. The present work aims to evaluate parameters at different cancer patients thawing. Testicular tumors lymphomas are among most common cancers in younger patients, while chemotherapy significantly increases patients' survival, it can epigenetically alter fluid, resulting temporary or permanent infertility. We analyzed data from database Gamete Cryopreservation Center (Annunziata Hospital, CS; Italy) period 2011-2020 cohort 254 aged 18-56 years. evaluation was blind manner anonymously recovered; main referring were assessed accordance with WHO guidelines decision limits (6th edition; 2021). types as follows: testis (TC; n = 135; 53.1%), hematological (HC; 76; 29.9%), other (OC; 43; 17%). Comparing TC vs. HC (P1) OC (P2), had worst quality: sperm number/mL (P1 0.0014; P2 0.004), total motility 0.02; 0.07), progressive 0.04; 0.05), viability 0.01; 0.02), percentage atypical morphology 0.05; 0.03). After thawing, recovery lowered, accounting for 46.82% 16.75%, respectively, whole cohort; similarly, subgroups ascribed TC, they showed lowest recovery. Strong correlation existed between pre- post-cryopreservation (p < 0.001) subgroup 0.05). All subgroups, extents, findings below reference values, suggesting diverse susceptibilities cryodamage. Cancer associated treatments affect quality, meaning should considered useful personalized prerogative timely manner.

Semen cryopreservation results in the differential remodeling of molecules presented sperm, and these alterations related to reductions sperm quality its physiological function have not been fully understood. Given this, this study aimed investigate cryoinjury mechanism goat by analyzing changes metabolic characteristics during process. The ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) technique was performed explore metabolite profiles fresh (C group), equilibrated (E frozen-thawed (F group). In total, 2570 metabolites positive mode 2306 negative were identified, respectively. After comparative analyses among three groups, 374 differentially abundant (DAMs) C vs. E, 291 DAMs F, 189 E F obtained mode; concurrently, 530 405 193 mode, significantly enriched various pathways, including 31 pathways 25 28 Among them, 65 across comparisons discovered, which may be tightly associated with function. Particularly, functional terms such as TCA cycle, biosynthesis unsaturated fatty acids, sphingolipid metabolism, glycine, serine threonine alpha-linolenic acid pyruvate well pivotal like ceramide, betaine, choline, fumaric acid, L-malic L-lactic focused on. conclusion, our research characterizes composition their induced process, offering a critical foundation for further exploring molecular mechanisms metabolism influencing freezing tolerance sperm. Additionally, impacts equilibration at low temperature on need more attentions compared thawing

Ovarian tissue cryopreservation (OTC) or testicular (TTC) are effective and often the only options for fertility preservation in female male patients due to oncological, medical, social aspects. While TTC resumption of spermatogenesis, either vivo vitro, has still be considered an experimental approach humans, OTC autotransplantation been applied increasingly preserve with more than 200 live births worldwide. However, reproductive cells followed by gametogenesis, may interfere sensitive highly regulated cellular processes. In particular, epigenetic profile, which includes not just reversible modifications DNA itself but also post-translational histone modifications, small non-coding RNAs, gene expression availability, storage related proteins transcripts, have this context. Due complex reprogramming maintenance mechanisms epigenome germ cells, growing embryos, offspring, carried out at very critical moments early life cycle. Given background, safety taking into account profile clarified. Cryopreservation mature (including Metaphase II oocytes spermatozoa collected via ejaculation invasively after biopsy) embryos used successfully many years Medically Assisted Reproduction (MAR). freezing vitro gametogenesis become attractive past, while few human studies analysed effects, most data deriving from animal studies. review, we highlight potential influence immature subsequent growth differentiation on humans animals.

Artificial insemination is an important assisted reproductive technology that has been applied in several mammalian species. However, successful cryopreservation of semen South American camelids limited, hindering the commercial application artificial alpaca In this scenario, addition antioxidants to extenders provides a strategy improve freezability sperm. Bioactive metabolites from natural extracts black maca have shown valuable antioxidant properties. Thus, objective study was evaluate effect atomized freezing medium epididymal spermatozoa alpacas. Fifteen pairs epididymis were collected local slaughterhouse. Each sample divided into six groups: (1) fresh, (2) yolk (YM), (3) 10 mg/mL maca, (4) 20 (5) 30 and (6) resveratrol (as control). Sperm performed through slow method. Markers associated with functionality, such as motility, viability, plasma membrane integrity, well markers oxidative damage, DNA total ROS production, mitochondrial function, analyzed. The results show supplementation (20 mg/mL) improved sperm function evaluated according index formazan deposits. Similarly, production decreased at mg/mL, although integrity did not any differences among groups. These suggest cytoprotective effects during freezing/thawing Further research will be focused on assessing by using biomolecular (proAKAP4) fertility.
