Abstract Cancer drug resistance poses a significant challenge in oncology, primarily driven by cancer cell plasticity, which promotes tumor initiation, progression, metastasis, and therapeutic evasion many different cancers. Breast cancers (BCs) are prominent example of that, with an estimated 2.3 million new cases 670,000 BC-related deaths registered worldwide annually. Triple-negative BC is especially challenging for treatments demonstrating particularly aggressive disease course, early manifestation metastatic disease, frequent drug-resistant types, poor individual outcomes. Although chemosensitizing agents have been developed, their clinical utility oncology remains unproven. The mitogen-activated protein kinase (MAPK) pathway considered critical regulator intracellular extracellular signaling highly relevant both — genetic epigenetic modifications. Dysregulation the MAPK pathways plays role conferring chemoresistance BC. Contextually, targeting represents promising strategy overcoming enhancing efficacy anticancer treatment. ABSTRACT Background Colon adenocarcinoma (COAD) is a leading cause of cancer‐related mortality, with limited therapies for advanced stages. Efferocytosis, the clearance apoptotic cells, modulates tumor immunity and progression. We investigated efferocytosis‐related genes (ERRGs) in COAD through multiomics integration. Methods analyzed data from public databases to identify differentially expressed ERRGs their molecular subtypes. An ERRG score index was developed using integrated machine learning algorithms evaluate its predictive capacity. Single‐cell sequencing vitro functional assays were performed validate key findings. Results Among 162 ERRGs, 22 dysregulated COAD. Three subtypes exhibited distinct prognoses, immune profiles, therapy responses. The system accurately predicted clinical outcomes, low scores correlating improved survival sensitivity certain drugs. analysis highlighted TIMP1 as regulator, confirmed by knockdown suppressing proliferation migration vitro. Conclusion demonstrate prognostic therapeutic relevance COAD, providing insights into subtyping immunotherapy prediction. emerges potential target, warranting further validation.

Impaired wound healing is one of the main clinical complications type 2 diabetes (T2D) and a major cause lower limb amputation. Diabetic wounds exhibit sustained inflammatory state, reducing inflammation crucial to diabetic management. Macrophages are key regulators in healing, their dysfunction would exacerbated poor wounds. Gene regulation caused by histone modifications can affect macrophage phenotype function during healing. Recent studies have revealed that targeting histone-modifying enzymes local, macrophage-specific manner reduce responses improve This article will review significance as well illustrate how polarization Targeting with may provide novel therapeutic strategies for treatment
