The skin serves as an essential barrier against pathogens and external insults, preventing moisture loss. Chronic conditions such atopic dermatitis stem from impairments in function. Circadian rhythms affect blood flow characteristics, which are significant for understanding dermatitis. Cinnamomum cassia bark, commonly known cinnamon, is extensively utilized both modern Traditional Chinese Medicine its therapeutic properties managing chronic diseases. This study aimed to investigate the potential use of bark enhancing We examined impact extract (CCBE) on circadian clock-mediated enhancement barrier. CCBE enhanced expression keratinocyte differentiation markers, including keratin 10, filaggrin, caspase 14, calpain-1. also increased production hyaluronic acid protein. Additionally, improved rhythm period regulator 2 ( PER2 ). Notably, upregulated markers specifically during morning hours. Furthermore, we discovered that siRNA-mediated knockdown diminished increase induced by CCBE. These findings demonstrate can regulate keratinocytes a time-of-day-dependent manner via clock. augmented phosphorylation p38 JNK, while CCBE-induced FLG was reduced MAPK inhibitors. results suggest strengthen diurnally pathway, representing novel more effective method function accommodates daily variations properties.

The recent emphasis on circadian rhythmicity in critical skin cell functions related to homeostasis, regeneration and aging has shed light the importance of PER2 clock gene as a vital antitumor gene. Furthermore, delta-opioid receptors (DOPrs) have been identified playing crucial role differentiation, proliferation migration, which are not only essential for wound healing but also contribute cancer development. In this study, we propose significant association between cutaneous opioid receptor (OPr) activity rhythmicity. To investigate link, conducted 48 h rhythm experiment, during RNA samples were collected every 5 h. We discovered that activation DOPr by its endogenous agonist Met-Enkephalin N/TERT-1 keratinocytes, synchronized dexamethasone, resulted statistically 5.6 delay expression core PER2. Confocal microscopy further confirmed simultaneous nuclear localization DOPr-β-arrestin-1 complex. Additionally, enhanced induced phase shift rhythmic binding β-arrestin-1 promoter. observed regulates transcription target genes, including PER2, facilitating histone-4 acetylation. Through ChIP assay, determined enhances acetylated H4 summary, our findings suggest leads via β-arrestin-1-facilitated chromatin remodeling. Consequently, these results indicate DOPr, much like healing, may play part development influencing

It is unclear whether normal human skin tissue or abnormal scarring are photoreceptive. Therefore, this study investigated photosensitivity in and hypertrophic scars. The expression of opsins, which photoreceptor proteins, dermal fibroblasts (NDFs) scar (HSFs) was examined. After exposure to blue light (BL), changes the levels αSMA clock-related genes, specifically PER2 BMAL1, were examined both fibroblast types. Opsins expressed types, with OPN3 exhibiting highest levels. peripheral circadian rhythm disruption, BL induced formation NDFs. In contrast, although HSFs showed gene levels, no distinct observed. level significantly higher decreased same as that NDFs upon exposure. When knocked-down exposed BL, reduction inhibited. This directly triggers synchronization but not HSFs. OPN3-mediated inhibited Although current results did elucidate relationship between rhythms scars, they show can be applied for prevention treatment scars keloids.

The pattern of itching in patients with atopic dermatitis has not been systematically studied. Therefore, this study aimed to assess the adults using questionnaires for a circadian rhythm participating at single institution (n = 241). A self-report questionnaire was used and intensity patients. In addition, patients' disease severity (Eczema Area Severity Index [EASI]) quality life (Dermatology Life Quality [DLQI]) were assessed. Itching occurred most frequently (74.69%) greatest (62.66%) between 20:00 00:00, least number (25.31%) experienced 04:00 08:00. DLQI EASI scores both correlated average maximum itch (r 0.582, r 0.533, respectively; 0.539, 0.517, p < 0.001). associated (B 0.179, B 0.204, 95% CI: 0.112 0.246, 0.096 0.313, 0.001), score males family history 0.285, 0.287, 0.094 0.476, 0.478, 0.003). Adult exhibited itching; these results could positively impact treatment approaches.
