Endothelial dysfunction is associated with the progression of sepsis. This study sought to probe molecular route sex-determining region on Y chromosome-box transcription factor 18 (SOX18) in sepsis-associated endothelial injury. Human umbilical vein cells (HUVECs) were treated lipopolysaccharide (LPS) establish sepsis cell model. Cell viability, lactate dehydrogenase (LDH) release, oxidative stress (reactive oxygen species/malondialdehyde/superoxide dismutase), and inflammation (interleukin-1β/tumor necrosis factor-α/interleukin-6) evaluated by counting kit-8 assay relevant kits. The expression levels SOX18, microRNA (miR)-204-5p, cadherin-2 (CDH2) determined real-time quantitative polymerase chain reaction Western blot assay. interaction miR-204-5p, CDH2 was analyzed chromatin immunoprecipitation dual-luciferase LPS induced HUVECs injury downregulation SOX18. SOX18 overexpression increased while decreased LDH activity, stress, inflammation. bound miR-204-5p promoter promote expression, further repressed expression. knockdown abrogated protective role Overall, alleviated LPS-induced promoting repressing CDH2, suggesting it as a potential target for treatment.
