Glioblastoma multiforme (GBM) is regarded as the most aggressive form of brain tumor delineated by high cellular heterogeneity; it resistant to conventional therapeutic regimens. In this study, anti-cancer potential garcinol, a naturally derived benzophenone, was assessed against GBM. During analysis, we observed reduction in viability rat glioblastoma C6 cells at concentration 30 µM extract (p < 0.001). Exposure garcinol also induced nuclear fragmentation and condensation, evidenced DAPI-stained photomicrographs cells. The dissipation mitochondrial membrane dose-dependent fashion linked activation caspases. Furthermore, that mediated inhibition NF-κB 0.001) decreased expression genes associated with cell survival (Bcl-XL, Bcl-2, survivin) proliferation (cyclin D1). Moreover, showed interaction through some important amino acid residues, such Pro275, Trp258, Glu225, Gly259 during molecular docking analysis. Hepatocellular carcinoma (HCC) is a tumor of high heterogeneity and complexity, which poses significant challenges to effective treatment patient prognosis because its immune evasion characteristics. To address these issues, single-cell technology machine learning methods have emerged as promising approach identify genes associated with escape in HCC. This study aimed develop prognostic risk score model for HCC by identifying intrinsic immune-evasion (IIEGs) through learning, providing insights into infiltration, enhancing predictive accuracy, facilitating the development more strategies. The utilized data from Cancer Genome Atlas database analyze gene expression profiles clinical related patients Various tools, including Human Protein Atlas, cBioPortal, analysis, Kaplan-Meier plot, were used IIEGs. Functional enrichment analysis was conducted explore potential mechanisms. In addition, abundance infiltrating cells microenvironment investigated using single-sample set CIBERSORT, xCELL, immunophenotype algorithms. glycosylphosphatidylinositol anchor attachment 1 (GPAA1) examined sample quantitative real-time polymerase chain reaction, Western blotting, immunohistochemical staining. Univariate Cox identified 63 IIEGs Using random forest, least absolute shrinkage selection operator regression support vector machine, consisting six (carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, dihydroorotase (CAD), phosphatidylinositol glycan biosynthesis class U (PIGU), endoplasmic reticulum membrane protein complex subunit 3 (EMC3), centrosomal 55 (CEP55), autophagy-related 10 (ATG10), GPAA1) developed, validated pairs adjacent non-cancerous samples. Based on calculated median score, samples categorized high- low-risk groups. curve showed that high-risk group had worse compared group. Time-dependent receiver operating characteristic demonstrated accurate capability prognosis. Furthermore, infiltration positive correlation between 40 checkpoint well Th2 cells. A formulated IIEG signatures promise predicting diagnosed utilization novel index, together significance valuable biomarker immunotherapy HCC, provides benefit determining therapeutic strategies application.

Glioblastoma multiforme (GBM), a highly aggressive and malignant form of brain cancer, continues to pose significant challenge in the field oncology. Despite ongoing advancements treatment strategies, prognosis for GBM patients remains grim, with 5-year survival rate hovering around 5%. The management involves multiple therapeutic approaches, including immunotherapy, but optimal outcomes terms overcoming tumor recurrence resistance have not been achieved. A key factor contributing therapy progression is tumor's ability evade immune system, referred as escape from cancer. This phenomenon reflects cells' efforts adapt survive body's response. release expression molecules like TGF-ß, IL-10, PD-L1, NKG2DL by cells impact activation, recognition, elimination system. Abstract Glioblastomas (GBM), the most common primary brain tumor, lack accurate prognostic markers and have a poor prognosis. Our study was designed to identify effective biomarkers for GBM prognosis analysis development of precise treatments. Differentially expressed genes (DEGs) between patients controls were analyzed from Xena database GEPIA. Based on screened DEGs, univariate COX LASSO regression performed relevant associated with Genes highly in selected construct receiver operating characteristic enrichment constructed groups high low expression adipocyte enhancer-binding protein 1 (AEBP1). CIBERSORT, ssGSEA ESTIMATE used perform immune infiltration analysis. About 3297 DEGs identified using data database; 8 identified. AEBP1, which plays role neuronal differentiation development, positively correlated GBMs infiltration; its cancer is short overall survival advanced tumor staging. This suggests that AEBP1 could serve as marker may better response immunotherapy.
