Abstract Truncated TrkB (TrkBT1), traditionally considered a dominant-negative regulator of full-length (TrkBTK+), remains poorly understood in peripheral sensory neurons, particularly nociceptors. Furthermore, neuronal expression and function has been associated with non-nociceptive Aδ low-threshold mechanoreceptors. This study challenges prevailing assumptions by demonstrating that TrkBT1 is the predominant isoform expressed neurons plays functional role modulating activity. We demonstrate nociceptors, identified markers such as TRPV1, TRPA1, TRPM8 5HT3A, well non-nociceptors, while (TrkBTK+) restricted to subpopulation. Functionally, we show acute application BDNF induces modest calcium influx nociceptors prolonged exposure significantly potentiates capsaicin-induced influx, an effect blocked TrkB-specific antagonist ANA12. Additionally, also promotes survival both nociceptive culture, dependent on Our data reveal ANA12 inhibits BDNF-mediated sensitization concentration-dependent manner, implicating distinct signaling pathways these processes. Collectively, our findings redefine modulator nociceptor activity rather than passive TrkB. By uncovering its dual roles survival, this provides new insights into molecular mechanisms BDNF/TrkB pain. Future work evaluating biology could offer perspectives how receptor contributes plasticity during chronic pain conditions.

Neurodegeneration, characterized by the progressive deterioration of neurons and glial cells, is a feature Alzheimer's disease (AD).
