TrkB
(TrkBT1),
traditionally
considered
a
dominant-negative
regulator
of
full-length
(TrkBTK+),
remains
poorly
understood
in
peripheral
sensory
neurons,
particularly
nociceptors.
Furthermore,
neuronal
expression
and
function
has
been
associated
with
non-nociceptive
Aδ
low-threshold
mechanoreceptors.
This
study
challenges
prevailing
assumptions
by
demonstrating
that
TrkBT1
is
the
predominant
isoform
expressed
neurons
plays
functional
role
modulating
activity.
We
demonstrate
nociceptors,
identified
markers
such
as
TRPV1,
TRPA1,
TRPM8
5HT3A,
well
non-nociceptors,
while
(TrkBTK+)
restricted
to
subpopulation.
Functionally,
we
show
acute
application
BDNF
induces
modest
calcium
influx
nociceptors
prolonged
exposure
significantly
potentiates
capsaicin-induced
influx,
an
effect

TrkB-specific
antagonist
ANA12.
