Chronic kidney disease (CKD) and diabetic (DKD) are major public health problems, their burden is growing relentlessly with the aging of global population. Their early recognition now a priority, there an unmet need for identification specific biomarkers in minimally invasive or non-invasive samples. Mitochondrial dysfunction plays pivotal role development progression both CKD DKD circulating platelets have emerged as ideal candidate assessment respiratory function. The present study assessed mitochondrial respiration isolated from peripheral blood patients compared to healthy controls. included total number 89 subjects, follows: 30 divided into three subgroups based on urinary albumin-to-creatinine ratio (uACR): 20 normoalbuminuric, 10 microalbuminuric, macroalbuminuric, 29 (positive controls) individuals (negative controls). Platelets were by differential centrifugations high-resolution respirometry protocol was adapted assess dependent complex I (CI) II (CII). A significant reduction CI-supported active found normoalbuminuric further decreased microalbuminuric subgroup. Both CI CII-dependent coupled maximal uncoupled significantly reduced macroalbuminuric In conclusion, impairment evident stages DKD. Moreover, platelet more severely impaired those CKD. A randomized, placebo-controlled trial was conducted assess the potential blood uric acid-lowering effects HRW consumption with different doses (low high doses) duration (4 8 weeks) in patients hyperuricemia. The Placebo group consumed three bottles ordinary drinking (330 mL per bottle), Low-HRW two bottle, H2 ≥ 4.66 mg/L) a bottle water, High-HRW daily for weeks. primary outcome acid levels following time points compared baseline. total 100 participants completed entire (32 Placebo, 35 Low-HRW, 33 groups). high-dose more effective than low-dose controlling acid. Following an 8-week period, exhibited significant reduction baseline (488.2 ± 54.1 μmol/L 446.8 57.1 μmol/L, P < 0.05). Metabolic disorders are a growing global concern, especially in developed countries, due to their increasing prevalence. Serum lipid profiles, including triglycerides (TG), total cholesterol (TC), high-density lipoprotein (HDL), and low-density (LDL), commonly used clinical biomarkers for monitoring the progression of these metabolic abnormalities. In recent decades, hydrogen-rich water (HRW) has gained attention as safe effective treatment, with regulatory effects on peroxidation inflammatory responses trials. This systematic review meta-analysis aim evaluate effectiveness HRW therapy blood profiles randomized controlled trials (RCTs) disorders. Following PRISMA guidelines, search RCT studies was conducted PubMed, Web Science, Embase, Google Scholar databases up January 2024. Eight that met all eligibility criteria, RCTs involving dysfunctions evaluations were included further analysis. Data extraction followed by quality evaluation using Jadad scoring system. Meta-analysis STATA software. The eight selected 357 patients various All showed either no or low risk bias. overall levels TG [95% CI: -0.27 (-0.47, -0.07)], TC -0.07 (-0.32, -0.18)], LDL -0.06 (-0.28, 0.15)] demonstrated slight decreases across studies. Coenzyme Q10 (CoQ10) is an essential medicinal ingredient. In this study, we obtained a high-yielding mutant strain of CoQ10, VK-2-3, by subjecting R. sphaeroides V-0 (V-0) to 12 C 6+ heavy ion beam and high-voltage prick electric field treatment. To investigate the mutation mechanism, complete genomes VK-2-3 were sequenced. Collinearity analysis revealed that nicotinamide adenine dinucleotide-dependent dehydrogenase (NAD) gene underwent rearrangement in genome. The NAD was overexpressed silenced V-0, construct named RS.NAD RS.ΔNAD. results showed titers CoQ10 RS.ΔNAD increased decreased 16.00 33.92%, respectively, compared those these differences significant. Our mechanism which yield increases through reverse metabolic engineering, providing insights for genetic breeding mechanistic analysis.
