Abstract Bladder cancer (BLCA) remains a widespread and serious globally. The importance of constructing predictive risk models based on DNA replication stress-related genes (DRSGs) is gaining traction. Despite the scarcity molecular studies, our analysis Cancer Genome Atlas (TCGA) Gene Expression Omnibus (GEO) datasets led to identification 17 DEDRSGs with prognostic significance, primarily involved in cell cycle control. Subsequently, utilizing LASSO regression, we filtered out nine key biomarkers develop model that effectively distinguishes immunological profiles GSVA hallmark pathways across strata. Besides, significant differences were observed distribution 191 drugs two groups. Finally, qRT-PCR was performed confirmed BUB1B significantly highly expressed 5637 cells line, while expression level RAD9A higher SV-HUC-1 line. In conclusion, model, biomarkers, offers theoretical framework for exploring underlying mechanisms BLCA.
