Resveratrol, a bioactive phytoalexin, has been extensively studied as pharmaceutical and nutraceutical candidate for the treatment of various diseases. Although its therapeutic effects have largely attributed to anti-oxidant properties, underlying mechanisms dose dependency are not well understood. Circular RNAs (circRNAs) are a class of circular non-coding that play essential roles in the intricate and dynamic networks governing cell growth, development, apoptosis. Resveratrol (RSV), non-flavonoid polyphenol, is known to participate follicular development ovulation. In our previous research, we established model using porcine ovarian granulosa cells (POGCs) treated with resveratrol, which confirmed its regulatory effects on long (lncRNAs) microRNAs (miRNAs) within these cells. However, influence resveratrol circRNA expression has not been thoroughly investigated. To explore how affects levels POGCs, designed an experiment three groups: control group (CON, n = 3, 0 μM RSV), low-dose RSV (LOW, 50 high-dose (HIGH, 100 RSV) for sequencing. We identified total 10,045 candidate circRNAs from POGCs different concentrations (0, 50, μM). Differential analysis indicated 96 were significantly altered LOW vs. CON group, while 109 showed significant changes HIGH group. These notably enriched biological processes associated metabolism, apoptosis, oxidative stress. Functional enrichment host genes revealed their involvement critical signaling pathways, including mTOR, AMPK, apoptosis pathways. Additionally, potential miRNA sponge candidates among differentially expressed circRNAs, particularly novel_circ_0012954 novel_circ_0004762, exhibited strong connectivity miRNA-target networks.
