Abstract Circulating tumor cells (CTCs) are cancer that shed from the primary and enter into body fluids of patient, where they travel to distant sites ultimately form metastasis. Understanding biology CTCs, in particular at critical stages their itinerary, holds promises for better cure. Since beginning this century, liquid biopsy has steadily grown be a keen area research due its non-invasive features. As one most promising biomarkers, CTCs have shown great potential diagnosis, prognosis, treatment response monitoring, exploration biological mechanisms. Although various types isolation detection technologies emerge constantly, rarity heterogeneity still pose huge challenges these methods make them inefficient. In addition, clinical practice different lacks reasonable uniform standards. review, we provide detailed overview enrichment strategies as well advantages limitations. Abstract Breast cancer (BC) threatens women’s health, and the prognosis is dismal. Folic acid metabolism affects prognosis, but research on folic metabolism-related genes (FMRs) in BC scarce. We used TCGA-BRCA as training set GSE21653 validation set. Five FMRs (PLAT, SERPINA3, IFNG, SLC19A1, NFKB2) were screened via univariate LASSO Cox regression analyses, a prognostic model was built based multivariate analysis. The showed excellent predictive performance. Differentially expressed high- low-risk groups enriched steroid hormone biosynthesis neuroactive ligand-receptor interaction pathways. group exhibited higher immune cell infiltration better immunotherapy response. AM-5992 5-fluorodeoxyuridine 10mer may be potential drugs. This FMR-based can accurately predict offering clinical reference.
