tRNA fragments (tRFs) are small non-coding RNAs generated from cleaved molecules, playing key roles in gene regulation and cellular processes. Produced by ribonucleases like angiogenin Dicer, tRFs vary length function silencing stress responses. They interact with Argonaute proteins affect mRNA levels, emerging as potential diagnostic therapeutic targets for diseases such cancer neurodegenerative disorders. Given that the skin is largest organ mammals, it serves an ideal model studying development various diseases. Therefore, this study investigates tRF expression sheep tissues to understand their regulatory during growth development. This analyzed tissue five 1-month-old lambs 24-month-old adult Tan using RNA sequencing proteomics. Raw data were filtered aligned identify tsRNAs, while proteomic assessed differential expression. Principal Component Analysis (PCA) revealed distinct separation between juvenile samples based on tsRNA patterns, indicating intra-group similarity inter-group differences. Differentially expressed tsRNAs identified, 19 highly at 1 month of age. Proteomic screening identified 932 835 lowly group, functional enrichment highlighting immunity inflammation pathways. Predictive analysis target genes intersected 20 differentially involved mitochondrial metabolism response. reveals significantly influence developmental immune processes sheep, patterns juveniles adults. Future research should validate these findings further elucidate mechanisms regulation.
