Macrophages are pivotal immune cells involved in maintaining homeostasis and defending against pathogens. They exhibit significant plasticity heterogeneity, enabling polarization into pro-inflammatory M1 or anti-inflammatory M2 phenotypes response to distinct microenvironmental cues. The process of macrophage is tightly regulated by complex signaling pathways transcriptional networks. This review explores the factors influencing polarization, associated pathways, their roles pathogenesis lung diseases, including fibrosis, cancer, chronic inflammatory conditions. By summarizing recent advances, we aim provide insights immunoregulatory functions macrophages therapeutic potential. Based on our review, it believed that targeting emerges as a promising approach for developing effective treatments balancing inflammation repair while mitigating disease progression.
