TGF-β is a pleiotropic cytokine with both stimulatory and inhibitory effects on immune cells, depending the microenvironmental context. Allergic contact dermatitis (ACD) is a skin condition characterized by inflammation resulting from hypersensitivity upon with certain allergens. Although ACD an immune-mediated pathomechanism, the involvement of nervous system in this has increasingly been considered, particularly amplification and persistence inflammation. This paper aims to present comprehensive overview mechanisms involved neurogenic ACD, focusing on role sensory neurons, release neuropeptides, their interaction immune cells, potential therapeutic implications related pathways, diversified age gender. Innovative therapies for including topical formulations, may target mass-bound X2 G-protein-coupled receptor (MRGPRX2) endocannabinoid systems.
