miR-211-5p exhibits dysregulated expression in various malignant tumors and has been implicated the regulation of tumor cell proliferation, apoptosis, inflammation, neurogenic processes. Previous studies have demonstrated that miR-211 negatively regulates ELOVL6, suggesting its involvement lipid metabolism lipogenesis within bovine adipose tissue. Our prior transcriptomic analysis revealed upregulated rabbits fed a high-fat diet, indicating potential role regulation. However, precise functions deposition rabbit preadipocytes remain unclear. To address this knowledge gap, study utilized as experimental models to investigate molecular mechanisms by which preadipocyte proliferation differentiation. The findings aim provide theoretical basis for improving meat quality. main are summarized follows: (1) EdU assay, RT-qPCR detection, CCK-8 viability assay overexpression inhibits preadipocytes, while inhibition promotes preadipocytes. (2) precursor adipocytes were transfected induced differentiate. RT-qPCR, western blot (WB), Oil Red O staining assays showed maturation differentiation rabbits, rabbits. (3) Through transcriptome sequencing, total 147 differentially expressed genes identified. Among them, TPK1 is target gene also newly identified important involved synthesis. (4) After silencing TPK1, series experiments, including WB, staining, conducted. results interfering with can inhibit promote their (5) co-transfection inhibitor si-TPK1, experiments such detection It was found targeting TPK1.
