Lead (Pb) is a heavy metal associated with range of toxic effects. Relatively few studies attempt to understand the impact lead on development from mechanistic perspective. Danio rerio (zebrafish) embryos are model organism for studying developmental consequences exposure chemical agents. This study examined metabolome developing zebrafish exposed 5 ppb, 15 150 and 1500 ppb Pb concentrations during first 24 h post fertilization, followed by unexposed harvest at 48 h. Untargeted metabolomics multivariate analysis revealed that various exposures differentially affected embryonic metabolome. Pathway analyses showed dysregulation biopterin, purine, alanine, aspartate metabolism. Inductively coupled plasma mass spectrometry demonstrated accumulation in embryos. Additionally, decreases oxidation–reduction ratios were observed 5–150 groups but not group. finding, along several metabolite abundances, suggests hormetic effect Together, these data reveal persistent global changes metabolome, pin-point biomarkers exposure, unveil dose-dependent relationships, reflect Pb-induced cellular energy. work highlights aberrant processes underlying low-dose early development.

Abstract Zebrafish ( Danio rerio ) have emerged as an influential model for studying human epithelial pathology, particularly because of their genetic similarity to humans and unique physiological traits. This review explores the structural functional homology between zebrafish tissues in organs, such gastrointestinal system, liver, kidneys. possess significant cellular with mammals, which facilitates investigation various diseases, including inflammatory bowel disease, nonalcoholic fatty liver polycystic kidney disease. The advantages using a organism include rapid external development, ease manipulation, advanced imaging capabilities, allowing real-time observation disease processes. However, limitations exist, concerning lack organs potential incomplete phenocopy conditions. Despite these challenges, ongoing research adult promises enhance our understanding mechanisms regenerative By revealing similarities differences cell function pathways, this highlights value translational advancing knowledge health developing targeted therapies.

The kidney is a vital organ responsible for removing toxins, producing urine, and regulating homeostasis. Developmental defects in the lead to various congenital abnormalities that impair renal function. Gdf11, member of transforming growth factor β family, associated with numerous abnormalities. In early developmental stage, pronephric duct hindgut open into cloaca, Gdf11 shows significant expression mice. However, molecular cellular roles gdf11 cloaca organogenesis remain unclear. Our study revealed pronephros formation were significantly disrupted upon deletion or knockdown zebrafish. Additionally, we found TGF-β pathway acts downstream promoting development. Treatment small molecule activator partially rescued observed gdf11-/- mutants. summary, our findings provide strong evidence critical link between pronephros/cloaca signaling mediated by Gdf11. also provides new insight diseases related

Recent years have heralded many exciting advancements in our knowledge about kidney development. In particular, there has been tremendous progress identifying genes and signaling pathways that pattern renal functional units - a process known as nephron segmentation. An intriguing potential regulator of this process, 17β-estradiol (E2), was implicated previously by high-throughput screen examined the effects bioactive molecules on nephrogenesis. Now, detailed study shown exogenous E2 or exposure to several xenoestrogens significant distal tubule establishment during development zebrafish pronephros, embryonic kidney. Attenuation estrogen receptor 2b (Esr2b) activity pharmacological antagonism genetic knockdown revealed E2/Esr2b is necessary for normal segment regulating expression Iroquois transcription factors. These findings demonstrate influences stem cell vertebrate segmentation may important ramifications understanding congenital birth defects diseases.

Kidney disease is a devastating condition affecting millions of people worldwide, where over 100,000 patients in the United States alone remain waiting for lifesaving organ transplant. Concomitant with surge personalized medicine, single-gene mutations, and polygenic risk alleles have been brought to forefront as core causes spectrum renal disorders. With increasing prevalence kidney disease, it imperative make substantial strides field genetics. Nephrons, functional units kidney, are epithelial tubules that act gatekeepers body homeostasis by absorbing secreting ions, water, small molecules filter blood. Each nephron contains series proximal distal segments explicit metabolic functions. The embryonic zebrafish provides an ideal platform systematically dissect genetic cues governing development. Here, we review use discover nephrogenesis genes.
