The paper aims to investigate the biological role of microRNAs secreted by preimplantation embryo into blastocoel fluid and detect a distinctive molecular signature for identifying embryos with highest implantation potential. We carried on multicenter retrospective study involving five European IVF centers. collected 112 samples from day 5 post-fertilization, cultured individually, along data blastocyst grade transfer outcomes. Using custom TLDA Array, we compared expression levels 89 miRNAs between 33 fluids high-quality implanted 30 not-implanted embryos. Expression differences were assessed using SAM t-test. Additionally, correlation function enrichment analysis network construction conducted identify roles deregulated microRNAs. identified six up-regulated in embryos, significantly positively correlated across all (r ≥ 0.7; P ≤ 0.05). They could take part pluripotency circuits, regulating being regulated transcription factors associated stemness, cell growth, development. ROC curve confirmed potential these as classifiers. may represent functional evaluating quality most competent Their evaluation non-invasive genetic testing, integrating epigenetic genomic analyses, enhance allow identification euploid not able implant.
