Septins can function as scaffolds for protein recruitment, membrane-bound diffusion barriers, or membrane curvature sensors. are important cytokinesis, but their exact roles still obscure. In fission yeast, four septins (Spn1 to Spn4) accumulate at the rim of division plane rings. The octameric exocyst complex, which tethers exocytic vesicles plasma membrane, exhibits a similar localization and is essential deposition during cytokinesis. Without septins, spreads across absent from septum formation. These results suggest that physically interact proper localization. Indeed, we predicted six pairs direct interactions between septin subunits by AlphaFold2 ColabFold, most them confirmed co-immunoprecipitation yeast two-hybrid assays. Exocyst mislocalization in mistargeting secretory cargos, leads cell-separation delay mutants. Our indicate guide targeting complex on vesicle tethering cytokinesis through physical interactions.
