Inonotus obliquus is a medicinal fungus that has been favored for its anti-diabetic benefits in China thousands of years. Inotodiol characteristic triterpenoid isolated from obliquus. The inactivation effect and mechanism α-glucosidase induced by inotodiol were investigated using fluorescence, FTIR, CD spectroscopy, molecular dynamics simulation. could effectively inhibit activity (IC50 = 1.09 ± 0.08 mmol/L) reversible noncompetitive manner. interacted with via static quenching process binding constant 1.88 × 103 mol/L at 298 K. Molecular simulation revealed mainly hydrogen bonding the complex system was stable. also increase glucose consumption glycogen synthesis insulin-resistant HepG2 cells through IRS-1/PI3K/AKT-mediated GSK-3β signaling pathway. Overall, this study suggested potential to be an agent or functional food diabetics.
