Hyperhomocysteinemia (HHcy) influences the development and progression of neurodegenerative disorders in different ways. Homocysteine (Hcy) metabolism is related to that asymmetric dimethylarginine (ADMA) group B vitamins. The breakdown pathway involving nitric oxide (NO) ADMA can be considered one causes endothelial alteration represents a crucial step several disorders. Deficiencies vitamins other than ones, such as D A, have also been associated with central nervous system aim this narrative review describe link between HHcy, ADMA, Parkinson’s disease (PD), Alzheimer’s (AD), multiple sclerosis (MS) terms dysfunctional pathways neuropathological processes, performing literature search from 2015 2025 on PubMed. This provides an overview effects vitamin supplementation diseases. NO production lead HHcy elevated concentrations, causing neurodegeneration through various mechanisms, while has shown reduce Hcy levels, although conflicting results about improvement clinical symptoms. Further studies are needed develop optimal combined therapeutic strategies.
