Background Carbapenem-resistant Acinetobacter baumannii (CRAB) poses a severe nosocomial threat, prompting need for efficient detection methods. Traditional approaches, such as bacterial culture and PCR, are time-consuming cumbersome. The CRISPR-based gene editing system offered potential approach point-of-care testing of CRAB. Methods We integrated recombinase polymerase amplification (RPA) CRISPR-Cas12a to swiftly diagnose CRAB-associated genes, OXA-51 OXA-23 . This multiplex RPA-CRISPR-Cas12a eliminates bulky instruments, ensuring simplified UV lamp-based outcome interpretation. Results Operating at 37°C 40°C, the entire process achieves CRAB diagnosis within 90 minutes. Detection limits genes 1.3 × 10 −6 ng/μL, exhibiting exclusive without cross-reactivity common pathogens. Notably, platform shows 100% concordance with PCR when 30 clinical strains. Conclusion In conclusion, our RPA coupled provides fast sensitive method, overcoming limitations traditional approaches holding promise testing.

The glycopeptides dalbavancin, telavancin, oritavancin highly bound plasma proteins. Although effective animal models meningitis, it is unlikely that they reach cerebrospinal fluid (CSF) concentrations after intravenous administration alone. β-lactam/β-lactamase inhibitor combinations have principal problem both compounds must achieve adequate CSF concentrations. In commercially available combinations, dose β-lactamase tends too low oxazolidinone tedizolid has broader spectrum but less suitable profile than linezolid. halogenated tetracycline eravacycline does not sufficient treat usual dosing. Acinetobacter (A.) baumannii is a major nosocomial pathogen in human and veterinary medicine. The emergence of certain international clones (ICs), often with multidrug-resistant (MDR) phenotypes biofilm formation (BF), facilitates its spread clinical environments. global rise antimicrobial resistance demands alternative treatment strategies, such as peptides (AMPs). In this study, 45 companion animal MDR-A. isolates, belonging to the globally IC1, IC2 IC7, were tested for biofilm-associated genes (BAGs) their capacity BF. Of these, 13 used test inhibitory effect AMPs on bacterial growth (BG) BF through application crystal violet assay. two novel AMP variants Bac7(17) (target cell inactivation) Pasmr5-17 (efflux pump inhibition) well-known phenylalanine-arginine-β-naphthylamide (PAβN) at concentrations 1.95 1000 µg/mL. Based whole-genome sequence data, identical patterns BAGs detected within same IC. inhibited BG dose-dependent manner. PAsmr5-17 highly effective against BG, inhibition (GI) >99% (62.5 125 µg/mL, respectively). PAβN achieved only 95.7% GI Similar results obtained Differences between ICs found both when influenced by AMPs. had hardly any IC1 but IC7 31.25 µg/mL was sufficient. Our data show that susceptibility most likely resembles depending assignment particular Even low significant BG. Therefore, represent promising baumannii, either sole therapy or combination antibiotics.
