Salmonella enterica serovar Infantis has emerged as a prevalent foodborne pathogen in poultry with significant global health implications. This study investigates the molecular characteristics influencing virulence S. rough variant collected from farm USA. In this study, whole genome sequencing and comparative genomics were performed on smooth isolates, while chicken embryo lethality assay was conducted to assess their virulence. Comparative between isolates analyzed using Mauve pairwise Locally Collinear Blocks measure genetic conservation. Embryo survival rates compared Kaplan–Meier curves. High genomic conservation observed two but frameshift mutation detected Wzz(fepE) gene of variant, resulting early protein truncation. The showed that rate strain higher than (p < 0.05). identifies gene, leading truncation, which may reduce bacterial by impacting O-antigen biosynthesis variant. These findings deepen our understanding pathogenesis suggest targeting or related pathways could be promising strategy for developing effective vaccines therapeutic interventions.
