For decades, amyloid β-peptide (Aβ) misfolding aggregates with β-sheet structures have been linked to the occurrence and advancement of Alzheimer’s disease (AD) development progression. As a result, modulating mode Aβ has regarded as an important anti-amyloid protein strategy. A polyoxometalate based on {Co(H2O)4}2+ complex [P2Mo5O23]6− fragments, K8{[Co(H2O)4][HP2Mo5O23]2}·8H2O (abbreviated CoPM), synthesized structurally characterized using elemental analysis, single-crystal X-ray diffraction (SXRD), IR, UV spectra, bond valence sums (Σs) calculation, powder XRD (PXRD). CoPM’s primary component, revealed by structural is nanoscale polyoxoanion made [Co(H2O)4]2+ sandwiched between two pieces. Notably, it demonstrated that CoPM efficiently modulates aggregates’ β-sheet-rich conformation.

Amyloid β-peptide (Aβ) misfolding aggregates with β-sheet structures and surplus reactive oxygen species (ROS) are both considered to be the culprit of neuronal toxicity in Alzheimer's disease (AD). Therefore, modulating mode Aβ inhibiting ROS simultaneous has become an important method for anti-AD. Herein, a nanoscale manganese-substituted polyphosphomolybdate (H2en)3[Mn(H2O)4][Mn(H2O)3]2[P2Mo5O23]2·14.5H2O (abbreviated as MnPM) (en = ethanediamine) was designed synthesized by single crystal transformation method. MnPM can modulate rich conformation aggregates, thus reduce formation toxic species. Moreover, also possesses ability eliminate free radicals produced Cu2+-Aβ aggregates. It inhibit cytotoxicity β-sheet-rich protect synapses PC12 cells. combines anti-oxidation ability, which makes promising multi-funcational molecular composite mechanism new conceptual designing treatment such protein-misfolding diseases.

We introduced a terminal alkyne into the core structure of dolutegravir, resulting in synthesis 34 novel dolutegravir-1,2,3-triazole compounds through click chemistry. These exhibited remarkable inhibitory activities against two hepatocellular carcinoma cell lines, Huh7 and HepG2. Notably, 5e 5p demonstrated exceptional efficacy, particularly cells, with IC50 values 2.64 5.42 μM. Additionally, both induced apoptosis suppressed tumor clone formation, elevated reactive oxygen species (ROS) levels, further promoting apoptosis. Furthermore, activated LC3 signaling pathway, inducing autophagy, triggered γ-H2AX DNA damage cells. Compound low toxicity, highlighting its potential as promising anti-tumor drug.
