Abstract A novel modified sensor based on electropolymerization of hippuric acid (HA) using cyclic voltammetry within the potential window − 1 to 1.5 V for 10 cycles at a scan rate 100 mV s −1 over multiwalled carbon nanotubes (MWCNTs) battery graphite electrode (BGE). Poly (HA)/MWCNTs/BGE exhibited two linearity ranges 3.00 × −3 1.00 µM (5.29 −4 – 0.18 µg/ml) and 5.00 3 (0.88− 176.22 with limit detection (LOD) 0.06 −2 (1.06 quantification (LOQ) 2.00 (3.52 µg/ml). Experimental designs are essential mathematical tools in fields like agronomy, chemistry, and analytical chemistry for optimizing processes minimizing variations. Doehlert designs, particular, valued their efficiency exploring experimental space with minimal experiments, providing detailed insights into complex processes. In these extensively used tasks such as extraction, purification, method optimization, allowing systematic variation of factors to enhance accuracy efficiency. To further optimize the method, a combination design chemometric is necessary understand chromatographic behavior compounds interest. This study uses combined elution separation neurotransmitters metabolites using HPLC-ECD. Pearson correlation Partial Least Squares Discriminant Analysis (PLS-DA) reveal significant relationships between parameters conditions. Notably, pH mobile phase significantly impacts column time, while polarity index pressure influence peak asymmetry. Optimized conditions include ACN/MeOH/H2O (11.25/3.25/85.5; v/v/v) near 1.65, achieving optimal times around 20 min, mean number theoretical plates close 8000, asymmetry approximately 1.23. The limits detection quantification interest 10-11 10-10 mol L-1 respectively. new approach allows effective, rapid, resolved compounds, reducing resource consumption time. Moreover, has been taken account chemometrics, highly effective enhancement compound elution. optimized achieves low nanomolar range, making it suitable precise neurotransmitter analysis biological samples.

Objective: Neuropathic pain (NP) arises from trauma to the somatosensory nervous system and can be managed using selective serotonin reuptake inhibitors, such as fluoxetine (FLX) phosphodiesterase sildenafil (SD), cholesterol-lowering agents lovastatin (LOVA). The present study aimed develop validate an analytical method for simultaneous estimation of these drugs (SD, FLX, LOVA [SFL]) reverse-phase high-performance liquid chromatography (RP-HPLC). Methods: An RP-HPLC was developed validated quantification SFL. Chromatographic separation achieved a C-18 ODS column with mobile phase consisting acetonitrile 0.2 M ammonium acetate buffer (55:45) in gradient elution mode. flow rate maintained at 1 mL/min, detection carried out 228 nm. following ICH Q2 (R2) guidelines, assessing parameters linearity, accuracy, precision, limit (LOD), (LOQ). Results: exhibited linearity within concentration range 20–100 μg/mL, regression coefficient (r2) 0.9992. Retention times SD, were recorded 6.481, 4.238, 19.778 min, respectively. Recovery studies demonstrated accuracy 94.61–110.44%, relative standard deviation 0.06–2.00%, confirming precision method. LOD values found 12.77 14.81 13.28 respectively, while LOQ 45.16 42.33 38.71 μg/mL. Conclusion: met all required validation criteria suitability accurate pharmaceutical formulations. These findings support potential use alternative therapeutic strategy NP.
