Background Hepatic encephalopathy (HE) is a neurological disorder resulting from advanced liver injury. HE has high mortality rate and poor prognosis. The pathogenesis of still unclear, which led to the lack satisfactory specific treatment method. There increasing evidence that intestinal flora affects communication between gut brain in HE. Adjusting had beneficial effect on recent studies, Qingchang Ligan formula (QCLG) been shown previous studies regulate metabolites. In this study, we established thioacetamide-induced mouse model evaluate protective QCLG explore its potential mechanism, also demonstrated dysbiosis involved Methods Mice were intraperitoneally injected with thioacetamide (TAA, 150 mg/kg) induce Additionally, they orally administered Formula at dose 6.725 g/kg·d for seven days, while control mice received an equal volume saline via gavage. Subsequently, samples subjected 16S ribosomal ribonucleic acid (rRNA) gene sequencing, high-performance liquid chromatography-mass spectrometry (LC-MS), RNA-sequencing (RNA-seq) analysis. Result improved weight loss, cognitive impairment, function scores, blood ammonia, expression interleukin-6 (TNF-α), Interleukin-1β (IL-1β), (IL-6) induced by Moreover, increased levels indicators, including alanine aminotransferase (ALT), aspartate (AST), serum TNF-α, IL-1β, IL-6. RNA sequencing revealed Oscillibacter, Colidextribacter , Helicobacter TAA-induced fecal samples. Also, abundance Bifidobacterium decreases contrast, significantly restored microbial community. Metabolomics indicated significant differences some metabolites among normal control, treatment, groups, 5-methoxytryptophan, Daidzein, Stercobilin, Plumieride (PLU). Conclusion can alleviate neuroinflammation prevent caused injury regulating models.

Cyclophosphamide (CPM) is a classical alkylating agent used in different cancer chemotherapy regimens and restricted due to severe adverse effects, including hepatotoxicity. Natural or plant-derived antioxidants such as capsaicin were utilized this study examine the hepatoprotective benefits against cyclophosphamide-induced The rats divided into five groups: normal control group, toxic group (CPM), an intraperitoneal injection of single dose 200 mg/kg b.w. on fourth day, pretreated with two doses CPS (10 mg 20 b.w.) orally for six consecutive days, administration day treatment. fifth was administered highest (20 days. After 24 h CPS, anesthetized, blood collected, serum enzyme toxicity evaluated. sampling euthanasia all animals, liver isolated further histopathological examination. results revealed that markers (AST, ALT, ALP, BLI) significantly increased after CPM administration, but subsequently restored treatment both doses. In addition, lipid peroxidation (MDA), inflammatory cytokines (IL-1β, TNF-α), apoptotic (Caspase-3) increased, antioxidant enzymes (GSH, CAT, SOD) decreased it re-established by However, effectively protected CPM-induced architects tissues. conclusion, attenuates hepatotoxicity via modulating oxidative stress, signals, cytokine pathway. Therefore, could play significant role supplement during patients.

Impaired cognition is the primary symptom of dementia, which can lead to functional disability and reduced quality life among an increasingly ageing population. Ageing associated with increased oxidative stress, chronic low-grade systemic inflammation, endothelial dysfunction, reduces cerebrovascular function leading cognitive decline. Chronic inflammatory conditions, such as obesity, exacerbate this decline beyond normal predispose individuals neurodegenerative diseases, dementia. Capsaicin, major pungent molecule chilli, has recently demonstrated improvements in animal models via activation transient receptor potential vanilloid channel 1 (TRPV1). Capsaicin-induced TRPV1 adiposity, well improves function, all are cognition. This review examines current literature on capsaicin Capsimax, a supplement gastrointestinal irritation compared capsaicin. Acute treatment improve animals. However, studies adequately assessing effects humans do not exist. Capsimax may be potentially safe therapeutic intervention for future clinical trials testing

Background/Objectives: Diabetes mellitus is often accompanied by mental health complications, including anxiety, depression, and cognitive decline. Recent research suggested that capsaicin, the active component of chili peppers, may influence health. This study aimed to determine effect dietary capsaicin on disorders in a type 1 diabetes (T1D) mouse model, while also exploring potential involvement microbiota-gut-brain axis. Methods: We induced T1D mice using streptozotocin (STZ) administered diet supplemented with 0.005% for five weeks. Behavioral assessments, open field test (OFT), tail suspension (TST), forced swimming (FST), elevated plus maze (EPM) test, Morris water (MWM) were conducted evaluate depressive anxiety-like behaviors as well function. Targeted untargeted metabolomics analyses performed assess neurotransmitter levels hippocampus serum metabolites, 16S rRNA sequencing was utilized analyze gut microbiota composition. Intestinal barriers determined western blot detection tight junction proteins ZO-1 occludin. Results: Dietary exacerbated anxiety depressive-like along declines mice. Capsaicin reduced diversity beneficial bacteria, broad-spectrum antibiotic treatment further intensified depression behaviors. Metabolomic analysis indicated disrupted metabolic pathways related tryptophan phenylalanine, leading decreased neuroprotective such kynurenic acid, hippurate, butyric acid. Additionally, diminished expression occludin, indicating increased intestinal permeability. Conclusions: aggravates disturbances diabetic mice, thereby worsening

ABSTRACT Atherosclerosis (AS) is the basis of cardiovascular diseases (CVDs) and remains major contributor to death worldwide. Capsiate derived from sweet pepper fruit exhibits numerous pharmacological activities. The objective this study was elucidate protective role capsiate in atherosclerosis by examining its effect underlying regulatory pathways. Here, we showed that treatment alleviates atherosclerosis-prone apolipoprotein E-deficient (ApoE −/− ) mice. We found effectively reduced plaque area body weight compared Model group. inhibited inflammatory response downregulating phosphoinositide 3-kinase/protein kinase B/nuclear factor-κB pathway. Additionally, further investigation indicated could regulate lipid levels mice via reducing expressions 3-hydroxy-3-methylglutaryl coenzyme A reductase low-density lipoprotein receptor, increasing expression recombinant cytochrome P450 7A1. Furthermore, activated transient receptor potential vanilloid subfamily member 1 ApoE fed a high-fat diet. microbial sequencing demonstrated administration significantly regulated gut microbiota disturbance increased some beneficial bacterial ( Lachnospiraceae NK4A136 group) Human umbilical vein endothelial cells (HUVECs) were exposed oxidized (ox-LDL) stimulate atherosclerotic damage vitro . Our revealed ox-LDL-induced HUVECs injury inflammation. investigated effects on ferroptosis vivo ; it exhibited anti-ferroptosis through regulating nuclear factor erythroid 2-related 2/glutathione peroxidase 4 Interestingly, ML385 reversed HUVECs. Taken together, our findings suggest promising use small-molecule drug for AS related CVDs. IMPORTANCE has been inhibit fat accumulation, promote energy metabolism, exhibit anti-inflammatory antioxidative properties. However, there still no mouse models. first report reshaping structure AS, explore mechanism improvement AS. In study, alleviate diet-induced inhibiting response, improving serum profiles, activating pathway, suppressing ferroptosis. Moreover, reported as mediators therapy animal Therefore, these may provide robust experimental support clinical treatment.
