ABSTRACT This study aims to extract pepsin soluble collagen (PSC) from sturgeon cartilage, hydrolyze cartilage peptides (SCCP), and prepare SCCP nanoliposomes explore the treatment effects of osteoarthritis (OA) in rats. PSC was extracted using 0.5 M acetic acid (10%) enzymatically hydrolyzed with 4.5% alcalase plus flavourzyme obtain SCCP. Amino analysis revealed presence glycine, proline, hydroxyproline high amounts, while SDS‐PAGE showed that belonged type II molecular weight (MW) being <2 kDa MALDI‐TOF‐MS indicated MW distribution range 302.594 683.050 Da peptide fragments <500 accounting for 89.71%. composed phosphatidylcholine, fatty sucrose ester, glycerol, deionized water were prepared size at 34.58 nm, polydispersity index 0.19, zeta potential ‐54.53 mV, encapsulation efficiency 88.14%. Tube feeding SCCP/SCCP into OA rats alleviated pain responses by joint damage through reduction hind limb weight‐bearing difference, knee width levels serum biomarkers including CTX‐II, TGF‐β1, PIICP, COMP. Histopathologic images demonstrated mitigation symptoms tissue reducing damage, inhibiting chondrocyte apoptosis, promoting regeneration, synovitis. Collectively, dose most effective alleviating possessing a great be developed health food or botanic drug joint‐related disease.

Objective Mounting evidence suggests that histone deacetylases (HDAC) inhibitors reduce cartilage destruction in animal models of osteoarthritis (OA). Tumor necrosis factor (TNF)-α-blocking treatment for OA may provide effective joint protection by slowing damage. To investigate the effects intraperitoneal administration etanercept (a TNF-α inhibitor) on development rats and changes nociceptive behavior expression HDACs, RUNX2, MMP13 cartilage. Methods Induction Wistar was accomplished through anterior cruciate ligament transection (ACLT). One or five milligrams (mg) administered intraperitoneally 5 consecutive weeks after ACLT to + (1 mg/kg) groups. Nociceptive knee width were analyzed. Cartilage evaluated histologically immunohistochemically. Results significantly improved mechanical allodynia weight-bearing distribution compared alone. In treated with etanercept, degeneration synovitis less pronounced than those rats. OA-affected also showed reduced HDAC 6, 7, RUNX-2, MMP-13 response but increased HDAC4. Conclusion Our study demonstrated therapy (1) attenuated rats, (2) nociception, (3) regulated chondrocyte metabolism, possibly inhibiting cell HDAC6 HDAC7, increasing HDAC4 expression. Long non-coding RNA (LncRNA), with transcripts over 200 nucleotides in length, play critical roles numerous biological functions and have emerged as significant players the pathogenesis of osteoarthritis (OA), an inflammatory condition traditionally viewed a degenerative joint disease. This review comprehensively examines influence LncRNA on processes driving OA progression, focusing their role regulating gene expression, cellular activities, pathways. Notably, LncRNAs such MALAT1, H19, HOTAIR are upregulated exacerbate milieu by modulating key signaling pathways like NF-κB, TGF-β/SMAD, Wnt/β-catenin. Conversely, MEG3 GAS5, which downregulated OA, show potential dampening responses protecting against cartilage degradation influencing miRNA interactions cytokine production. By enhancing our understanding LncRNA' inflammation, we can better leverage them biomarkers for disease develop innovative therapeutic strategies management. paper aims to delineate mechanisms propose novel targets intervention.
