Neuropathic pain, a common symptom of several disorders, exerts substantial socioeconomic burden worldwide. Transient receptor potential vanilloid 1 (TRPV1), non-selective cation channel predominantly ex-pressed in nociceptive neurons, plays pivotal role nociception, by detecting various endogenous and exogenous stimuli, including heat, pro-inflammatory mediators, physical stressors. Dysregulation TRPV1 signaling further contributes to the pathophysiology neuropathic pain. Therefore, targeting is promising strategy for developing novel analgesics with improved efficacy safety profiles. Several pharmacological approaches modulate activity, agonists, antagonists, biological RNA interference (RNAi, small interfering [siRNA]) have been explored. Despite preclinical success, clinical translation TRPV1-targeted therapies has encountered challenges, hyperthermia, hypothermia, pungency, desensitization. Nevertheless, ongoing research efforts aim refine interventions through structural modifications, development selective modulators, discovery natural, peptide-based drug candidates. Herein, we provide guidance researchers clinicians involved new specifically reviewing existing literature highlighting current activities. This study discusses future endeavors enhancing efficacy, safety, tolerability candidates, thereby facilitates these discoveries into effective alleviate pain disorders.

The pathophysiology of fibromyalgia, a condition that causes chronic pain throughout the body, involves abnormal signaling, genetic predispositions, and neuroendocrine function, significantly impairing quality life. Fibromyalgia is commonly characterized by musculoskeletal pain, fatigue, severe sleep alterations. Changes in central processing sensory input defects endogenous inhibition could be basis enhanced persistent sensitivity individuals with fibromyalgia. term syndrome was chosen as an umbrella for fibromyalgia related illnesses, including myalgic encephalomyelitis/chronic fatigue syndrome, migraine, irritable bowel syndrome. Given substantial impact on health, there need new prevention treatment strategies, particularly those involving bioavailable nutraceuticals and/or phytochemicals. This approach important considering adverse effects current pharmaceutical treatments, such antidepressants anticonvulsants, which can lead to physical dependence tolerance. Natural products have recently been considered design innovative analgesics antinociceptive agents manage pain. Polyphenols show promise management neuropathic especially how anti-inflammatory corticosteroids nonsteroidal medical drugs, are effective only when inflammatory processes coexist not recommended primary

Introduction and purpose: Chronic pain, defined as pain persisting for three months or longer neuropathic which is a type that arises result of damage disease to the somatosensory system, including peripheral fibers (Aβ, Aδ, C fibers) central neurons, are serious conditions pose significant challenges modern medicine. Addressing these often necessitates multimodal approach analgesia, involving use multiple analgesic agents with different mechanisms action. Among capsaicin, naturally occurring compound found in hot peppers. This study will collect analyze information about applications capsaicin management, its action, efficacy, safety Brief description state knowledge: Capsaicin natural chili alkaloid highly selective agonist TRPV1 vanilloid receptor. Upon activation receptors, skin nociceptors become less sensitive stimuli. Incorporating into standard chronic management may enhance, expedite, streamline complex process treatment, ultimately contributing improved quality life patients communities. It also offers certain benefits treatment obesity, cardiovascular gastrointestinal pathologies, various types cancers, neurogenic bladder, some dermatological conditions. Materials Methods: Review summary research studies available databases on Google Scholar PubMed. Summary: Topically applied especially form an 8% patch valuable option treating bone joint pain. The highest evidence has been presented postherpetic neuralgia HIV-related where efficacy comparable treatment.
