Recent advancements have highlighted the potential of cannabis and its phytocannabinoids (pCBs) in skin health applications. These compounds, through their interaction with endocannabinoid system (ECS), show promise for products. Their ability to regulate inflammation, oxidative stress cell proliferation makes them useful addressing problems such as scarring, healing, acne aging, positioning valuable tools innovative skincare solutions. In present work, cellular molecular effects proprietary pCB-based formulations on ECS modulation, inflammation regeneration were investigated. Using human dermal fibroblasts (HDF) keratinocytes (HaCaT), effect both pre-treatment treatment scenarios following exposure stress-inducing agents was assessed. Key markers analyzed tackle efficacy mitigating promoting structural integrity regeneration. vitro results showed that these significantly reduced promoted improved functions. vivo studies confirmed well-tolerated led noticeable improvements health, including enhanced barrier function. This study demonstrates safety cosmeceutical By combining analysis testing, this research provides new insights into therapeutic pCBs managing various conditions.

Cannabidiol (CBD), one of the main actives from Cannabis sativa has been perpetually explored lately for its therapeutic effects. Its attributes, such as anti-inflammatory and antioxidant effects, snowball into pain management, epilepsy seizure alleviation, anxiety relief, well numerous other implications through entire metabolism. However, conventional administration routes challenge potential, with reported poor water solubility, hepatic degradation, gastric instability erratic bioavailability observed in oral administration. As a result, transdermal delivery systems have emerged promising alternative to or inhaled routes, offering improved targeted The medical use CBD throughout Europe, UK, USA Australia is extensive usually represented by pharmaceutical preparations recommended after treatment routs fail. non-medical limited each country's own legislation, wider range products being available, but irregular regulatory landscape coupled growing market cannabinoid-infused products, emphasizes need standardized formulations further clinical research. present work critically examines cannabidiol, explores skin's potential route strategies involved using it systemic targeting. We highlighted key challenges provided insights CBD`s variable based on different methods, thus compiling literature-based absorption, distribution, metabolism, excretion (ADME) study. also explore role endocannabinoid system, function various conditions, effects associated CBD, particularly light varying legislation across countries. Background: Cannabis sativa L. (C. sativa) has a long history of medicinal use. Its inflorescences contain bioactive compounds like non-psychotropic cannabidiol (CBD), which is well known for its anti-inflammatory potential in skin conditions such as psoriasis, and psychotropic Δ-9-tetrahydrocannabinol (THC). Keratinocytes, the main cells epidermis, are crucial regulating inflammation by producing mediators IL-8 when stimulated agents TNFα. Methods: This study explores effects standardized C. extract (CSE) with 5% CBD less than 0.2% THC human keratinocytes challenged The aim this to analyze specific contributions constituents CSE inflammatory responses fractionating examining individual components. Results: MTT assays showed that was non-toxic HaCaT up 50 μg/mL. inhibited NF-κB activity reduced secretion concentration-dependent manner, mean IC50 values 28.94 ± 10.40 μg/mL 20.06 2.78 (mean SEM), respectively. Fractionation into four subfractions revealed more lipophilic fractions (A B) were most effective inhibiting NF-κB, indicating cannabinoids cannflavins key contributors. Pure one active reducing NF-κB-driven transcription (together cannabigerol), due abundance CSE, it primarily responsible activity. Conclusions: highlights CBD’s significant role underscores need consider synergistic interactions several molecules within extracts maximum efficacy.
