Among neurotrophins, including nerve growth factor (NGF), brain-derived neurotrophic (BDNF), neurotrophin-3 (NT-3), and neurotrophin-4 (NT-4/5), BDNF has been extensively studied for its physiological role in cell survival synaptic regulation the central nervous system's (CNS's) neurons. binds to TrkB (a tyrosine kinase) with high affinity, resulting downstream intracellular signaling cascades play crucial roles determining fate, neuronal differentiation maturation of CNS It well demonstrated that downregulation/dysregulation BDNF/TrkB system is implicated pathogenesis neurologic psychiatric disorders, such as Alzheimer's disease (AD) depression. Interestingly, effects mimetic compounds flavonoids, small molecules which can activate TrkB-mediated signaling, have investigated potential therapeutic strategies brain diseases, given p75NTR, a common neurotrophin receptor, also contributes death under variety pathological conditions neurodegeneration. Since downregulation associated pathophysiology neurodegenerative diseases understanding how alterations contribute progression could provide valuable insight prevention these diseases. The present review shows recent advances molecular mechanisms underlying plasticity, providing critical insights into impact mimetics

This review explores the antidepressant effects of ginsenoside Rb1, a natural compound in traditional Chinese medicine, and its potential for treating major depressive disorder (MDD). The aetiology depression was reviewed up to 2024, focusing on pathways mechanisms through which Rb1 may exert effects. Notably, regulates oxidative stress inflammatory processes while enhancing neural plasticity by downregulating miR-134 expression alleviating symptoms. Unlike antidepressants that act single target, interacts with multiple pathways, reflecting broader therapeutic application. To compensate current deficiency animal experiments, clinical data, research side treatment depression, we some data use this component other diseases explore relevance depression. Ginsenoside is expected serve as novel or complementary combination compounds. However, further trials molecular studies are necessary confirm efficacy

Atherosclerosis (AS)-related cardiovascular disease and depression are often comorbid, with patients facing an increased risk of depression, which worsens AS. Both diseases characterized by oxidative stress lipid metabolism disorders. Ferroptosis, a form cell death iron overload harmful peroxide accumulation, is found in various diseases, including AS depression. Consistent the deposition peroxidation (LPO) that characterize ferroptosis mechanism, disturbances also crucial pathogenic mechanisms The comorbid complex, posing challenges for clinical treatment. Chinese herbs hold significant potential owing to their multi-target pharmacological effects. Therefore, this review aims investigate overload, LPO, across types, shared pathogenesis ferroptosis, research on herbal medicine targeting treatment anti-AS co-depression. This provides comprehensive understanding co-depression from perspective ferroptosis.
