The present study aimed to evaluate the effect of gabapentin-loperamide co-administration on induced diabetic neuropathic pain in female albino mice. involved 30 mice, divided into six groups: Group A served as negative group and received distilled water; B positive (induced pain); for 14 days, C included mice that solely oral gabapentin (2.5 mg/kg); D 2.5 mg/kg days orally loperamide 0.05 orally. E includes treated with 1.8 orally, F 1.2 (0.05 mg/kg) Results illustrated showed a significant decrease levels substance-p interleukin-6 (10.92 ±0.74 165.8 ±8.53, respectively) comparison those (108.78 ±4.46 1083.34 ±76.35, respectively), which may be attributed increase analgesic due reduction its elimination from body by decreasing small intestine movement increasing absorption, led keeping it long time.

Epilepsy is a neurological disorder characterized by repeated convulsions. Antiepileptic drugs (AEDs) are the main course of therapy for epilepsy. These medications given according to each patient’s personal medical history and types seizures they suffer. They have been employed decades manage epilepsy, thus delivering relief from through numerous mechanisms action. Aside their anticonvulsant attributes, current evidence suggests that certain AEDs may display potential inhibitory effects against cancer invasion metastasis. This review explored complicated interactions between modes action pathways causing cancer, impact on metastasis various forms while addressing associated side effects. For example, valproic acid inhibits histone deacetylase, hyperacetylation genes, especially those regulating cell cycle, culminating in cycle arrest. Topiramate carbonic anhydrase, disrupting acidic microenvironment needed cells thrive. Lacosamide increases slow inactivation voltage gated Na+ channel, inhibiting growth, proliferation, many cancers. Although drug development complex task due regulatory, intellectual property, economic challenges, researchers exploring repurposing tactics overcome these challenges find new therapeutic alternatives diseases like cancer. Thus, considered among most effective ways develop candidates using novel properties characteristics, this also discusses issues.

Introduction This study aimed to prepare cholesterol and stearic acid-based solid lipid nanoparticles of gabapentin (GAB-SLNs) for protection against streptozotocin (STZ)-induced retinal injury in rats. Methods We prepared four preparations GAB-SLNs using a hot high-shear homogenization ultrasonication process, the best formulation was selected tested biological activity. The brought male adult albino rats while doses continued 6 weeks. Six groups were assigned as vehicle, diabetic, diabetic + (10–20 mg/kg), mg/kg). GAB-SLN#2 optimized with high entrapment efficacy (EE%, 98.64% ± 1.97%), small particle size (185.65 2.41 nm), negative Zeta potential (−32.18 0.98 mV), low polydispersity index (0.28 0.02), elevated drug release (99.27% 3.48%). TEM image revealed smooth surface spherical shape. Results provided greater than free indicated by histopathology data which demonstrated more organization layers less degeneration ganglion cell layer treated GAB-SLN#2. Further, reduced inflammatory proteins (IL-6/JAK2/STAT3) vascular endothelial growth factor (VEGF). Conclusion preparation enhanced physical properties improved its activity neuroprotectant. Further studies are warranted validate this technique use oral other neurological disorders.
