Both hydrogen sulfide and endocannabinoids can protect the neural retina from toxic insults under in vitro vivo conditions. Purpose: The aim of present study was two-fold: (a) to examine neuroprotective action cannabinoids [methanandamide 2-arachidonyl glycerol (2-AG)] against peroxide (H2O2)-induced oxidative damage isolated bovine (b) evaluate role endogenously biosynthesized (H2S) inhibitory actions on stress retina. Methods: Isolated retinas cows were exposed using H2O2 (100 µM) for 10 min. When used, tissues pretreated with methanandamide (1 nM-100 nM) 2-AG (1-10 30 min before a treatment µM). In some experiments, inhibitors biosynthesis H2S [cystathionine β-synthase/cystathionine γ-lyase (CBS/CSE), aminooxyacetic acid, AOAA µM, or 3-mercaptopyruvate sulfurtransferase (3MST), α-keto-butyric KBA 1 mM] CB1-receptor antagonist, AM251 Enzyme immunoassay measurement 8-epi PGF2α (8-isoprostane) levels performed assess lipid peroxidation retinal tissues. Results: presence µM), significantly (p < 0.001) blocked H2O2-induced elevation 8-isoprostane CB1 antagonist nM), effect production attenuated. While (30 had no significant > 0.05) inhibition elicited by methanandamide, mM) reversed methanandamide. Conclusions: cannabinoids, prevent are partially dependent upon activation receptors endogenous via 3-MST/CAT pathway.
