Colchicine (COL), a widely used natural drug, has potent anti-inflammatory effects; however, as narrow therapeutic index its clinical application is limited by serious gastrointestinal adverse effects, and only oral formulations are currently marketed worldwide. Recent studies have shown that transdermal, injection, drug delivery the three main strategies for COL. This article elaborates on research progress of different in terms toxicity reduction efficacy enhancement, depicting transdermal route can avoid first-pass effect traumatic pain associated with injection routes, respectively. Microneedling, also known as percutaneous collagen induction, using microneedling devices and fabricated microneedle patches, has been widely employed in cosmetic applications for acne scar treatment, skin care, hair loss, melasma, rejuvenation, cancer. The micro-channels formed by through the stratum corneum facilitate delivery of agents stimulate elastin production inducing wound-healing cascade, keeping shiny wrinkle-free. Several agents, such ascorbic acid, hyaluronic retinoids, niacinamide, peptides, have delivered microneedling. This review aims to highlight use patches facilitating layers. Moreover, differences between devices, commonly used alone or combinational treatments with topical formulations, are explored. Predictive modeling of skin permeability for molecules: Investigating FDA-approved drug permeability with various AI algorithms DOI Creative Commons

The transdermal route of drug administration has gained popularity for its convenience and bypassing the first-pass metabolism. Accurate skin permeability prediction is crucial successful delivery (TDD). In this study, we address critical need to enhance TDD. A dataset comprising 441 records 140 molecules with diverse LogKp values was characterized. descriptor calculation yielded 145 relevant descriptors. Machine learning models, including MLR, RF, XGBoost, CatBoost, LGBM, ANN, were employed regression analysis. Notably, gradient boosting models outperformed others, demonstrating superior predictive accuracy. Key descriptors influencing permeability, such as hydrophobicity, hydrogen bond donors, acceptors, topological polar surface area, identified visualized. Cluster analysis applied FDA-approved (2326 compounds) revealed four distinct clusters significant differences in molecular characteristics. Predicted these offered insights into variations among drugs. Furthermore, an investigation patterns across 83 classes drugs based on ATC code showcased differences, providing valuable information development strategies. study underscores importance accurate TDD, emphasizing performance nonlinear machine models. key contribute a nuanced understanding characteristics These findings offer actionable design, formulation, prioritization optimum properties, potentially reducing reliance costly experimental testing.
