Benzo[a]pyrene (B[a]P) is known to inhibit neurodifferentiation and induce neurodegeneration. Agarwood or Aquilaria crassna (AC), a plant with health-promoting properties, may counteract the neurotoxic effects of B[a]P by promoting neuronal growth survival. This study investigated protective effect AC leaf ethanolic extract (ACEE) on B[a]P-induced impairment differentiation. A transcriptomic analysis identified canonical pathway, biological network, differentially expressed genes (DEGs) that are changed in response differentiation neurogenesis. Several genes, including CXCR4, ENPP2, GAP43, GFRA2, NELL2, NFASC, NSG2, NGB, BASP1, NEUROD1, B[a]P-treated SH-SY5Y cells were up-regulated after treatment ACEE. Notably, Western blot further confirmed ACEE increased protein levels GAP43 neuroglobin. led decreased phosphorylation Akt ERK cells; however, was able reverse these effects. Catechol-O-methyltransferase inhibitors (iCOMT), such as entacapone, have been successfully employed to treat tremor-related symptoms of Parkinson's disease. However, iCOMT has associated with a short half-life and poor oral bioavailability. Nanobased drug delivery systems often used overcome this type setbacks. Therefore, entacapone was encapsulated in PEGylated poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles (NPs) via nanoprecipitation process, well nanostructured lipid carriers (NLCs) using solvent emulsification/evaporation method. Both nanoformulations presented sub-200 nm populations, zeta-potential (ZP) values close -30 mV, showed stability at different pHs, while maintaining their physicochemical properties mostly intact, presenting only change superficial charge (ZP values), indicating interaction. interaction mucins, which anticipates good permeation bioavailability for topical administration.
