Brain diseases pose significant treatment challenges due to the restrictive nature of blood–brain barrier (BBB). Recent advances in targeting macromolecules offer promising avenues for overcoming these obstacles through receptor-mediated transcytosis (RMT). We summarize current progress brain drug delivery with diseases. This exploration details transport mechanisms across BBB, focusing on RMT and its use natural ligands delivery. Furthermore, review examines macromolecular such as antibodies, peptides, aptamers that leverage effective BBB traversal. Advancements macromolecules-based systems are summarized, emphasizing their therapeutic potential limitations. Finally, emerging strategies, including viral vectors, exosomes, boron neutron capture therapy, discussed precision brain-targeted treatments. comprehensive overview underscores RMT-based approaches revolutionize disease therapy.

Microglia, the resident immune cells of central nervous system, continuously monitor brain’s microenvironment through their array specific receptors. Once brain function is altered, microglia are recruited to sites perform functions, including phagocytosis misfolded proteins, cellular debris, and apoptotic maintain homeostasis. When toxic substances overproduced, over-activated produce large amounts pro-inflammatory cytokines, which induce chronic inflammatory responses lead neurotoxicity. Additionally, can also protect neuronal microglia-neuron crosstalk. Microglia receptors important mediators for receive external stimuli, regulate functional state microglia, transmit signals between cells. In this paper, we first review role microglia-expressed in pathogenesis treatment Alzheimer’s disease; moreover, emphasize complexity targeting therapeutic interventions neurodegenerative disorders inform discovery new biomarkers development innovative therapeutics

Introduction The blood-brain barrier (BBB) is a vascular endothelial membrane which restricts entry of toxins, cells and microorganisms into the brain. At same time, BBB supplies brain with nutrients, key substrates for DNA RNA synthesis, regulatory molecules, removes metabolic waste products from to blood. breakdown and/or dysfunction have been shown in neurogenerative disorders including Alzheimer's disease (AD). Current data suggests that these changes may initiate contribute neuronal, synaptic cognitive dysfunction, possibly other aspects neurodegenerative processes.

Background Chronic alcohol drinking is a modifiable risk factor for Alzheimer’s disease (AD), but underlying mechanisms remain poorly understood. Most studies of feeding to AD mice have utilized young and delivered in water without controlling nutritional intake. Objective To study the impact Lieber-DeCarli (LDC) liquid diet, which balances intake, on pathology aged Tg2576 wild-type (WT) mice, unexplored. Methods 13-month-old male female or WT were fed LDC diet (5% ethanol control) six weeks ( n = 11-13/group). Exploration (open-field test) spatial reference memory (Y-maze assessed after weeks, brains livers studied Aβ levels, synthesis transport proteins (APP LRP-1). Lactoferrin (Lf) is a multifunctional glycoprotein with well-established antimicrobial, anti-inflammatory, and iron-binding properties. Emerging evidence suggests that Lf also plays neuroprotective role, particularly in neurodegenerative disorders characterized by protein aggregation, such as Parkinson’s disease (PD). Alpha-synuclein (aSyn) aggregation pathological hallmark of PD other synucleinopathies, contributing to neuronal dysfunction progression. Recent studies indicate may interfere aSyn iron chelation, modulation oxidative stress neuroinflammation. Additionally, Lf’s ability cross the blood-brain barrier its potential impact on gut-brain axis highlight promise therapeutic agent. This review explores mechanisms action disease-modifying therapy, innovative delivery strategies could enhance clinical applicability. By addressing dimensions we propose compelling candidate for future research development diseases.
