In recent years, the bioactive compounds extracted from Sanghuangporus, especially polysaccharides, phenols, and triterpenoids, have attracted great interest people due to their extensive biological activity. Among them, polysaccharides are mainly seed bodies, mycelium, fermentation broth of Sanghuangyuan, exhibiting notable effects including immunomodulation, antitumor properties, hypoglycemic effects. This study employed ultrasound-assisted extraction (UAE) to isolate polysaccharides from Lyophyllum decastes, which were subsequently fractionated into two components, LDP-A1 and LDP-B1, using DEAE cellulose-52 Sephacryl S-500. The structural characteristics of the preliminarily analyzed high-performance liquid chromatography (HPLC), Fourier-transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), X-ray diffraction (XRD), Congo red staining. results indicate significant differences between LDP-B1 in terms molecular weight, monosaccharide composition, features. (2.27 × 106 Da) exhibits a significantly higher weight compared (9.80 105 Da), with distinct types content. Both contain β-glycosidic bonds. adopts sheet-like structure an amorphous internal arrangement triple-helix configuration, whereas is rod-shaped, crystalline structure, lacks configuration. In biological activity, both exhibit certain activities, but shows stronger activity antioxidant, hypoglycemic, anti-inflammatory, anticancer effects. summary, LDPs especially outstanding performance effects, proving their potential for development functional foods pharmaceuticals. ABSTRACT The goal of this study was to establish an efficient protocol for producing bioactive‐rich mushroom extracts food ingredients development. For this, fresh mushrooms were dried, ground, and submitted different extraction protocols—ethanolic (EE), fermentation‐assisted (FAE), or enzyme‐assisted (EAE). In addition, ultraviolet B (UV‐B) irradiation, coupled with the protocols, experimentally studied potential improvement bioactivity yields. yield (EY), (total phenolic content TPC, acids, flavonoids, antioxidant activity), α‐, β‐, total glucan contents, mineral evaluated. EAE yielded highest EY (26.48%–31.30%) while EE resulted in activity ( p < 0.05). β‐glucan (0.13%–0.93%) not affected by type UV‐B irradiation > All rich essential minerals low toxicity.
