Objectives: This study aimed to develop and validate a sensitive reliable liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the quantification of flualprazolam isotonitazene in serum samples address critical gaps forensic clinical toxicology. Materials Methods: A single-center validation was conducted using spiked with analyte standards. Analytical parameters, including linearity, precision, recovery, limit detection (LOD), (LOQ), were evaluated accordance international guidelines. The LC-MS/MS employed Shimadzu Triple Quadrupole™ MS 8045 system solid-phase extraction (SPE) sample preparation. Results: exhibited high linearity (r2 = 0.997) 0.999) over concentration range 1–100 ng/mL. LODs determined as 0.608 ng/mL 0.192 ng/mL, LOQs 1.842 0.584 isotonitazene, respectively. Recovery tests yielded results within acceptable 70–120%. Flualprazolam demonstrated recovery rates 98.0% 97.0% at theoretical concentrations 10 50 In contrast, slightly lower, measuring 75.5% 71.9% suggesting minor matrix effects that could influence its quantification. Precision analysis, both repeatability reproducibility, highlighted reliability method. %RSD values consistently below 7.07%, mean closely aligning across fortification levels. For remained 6.24%, although recoveries higher indicated potential challenges interaction. Conclusions: validated offers sensitivity, detecting serum, filling need toxicological investigations.
