Aflatoxin B1 (AFB1), a potent mycotoxin, poses significant threat to the poultry industry, particularly affecting health and growth of ducklings. The present study aimed investigate therapeutic effects mechanisms Tianjihuang compound (HRS), traditional Chinese medicine formulation, on AFB1-induced chronic toxicity in Firstly, 30 ingredients, including neochlorogenic acid, kaempferol 3-alpha-D-galactoside, quercetin, hispidulin, caffeic myricetin, were identified from HRS with UPLC-MS/MS method. Then, over 25-day experimental period, total 100 one-day-old Sichuan Sheldrakes randomly divided into five groups: control, AFB1 model, high (4 g/kg), medium (2 low dosage (1 g/kg) groups. Results indicated that effectively mitigated negative impact productivity, reduced levels liver index, AST, ALT, AST/ALT serum, increased serum TP content, obviously alleviated inflammatory cell infiltration, fibrosis, steatosis induced by AFB1. Additionally, enhanced GST, CAT, T-AOC, decreased MDA AFB1-DNA, thereby alleviating oxidative stress AFB1-DNA generation caused Transcriptome analysis revealed may improve injury AFB1-chronically poisoned ducklings regulating ECM receptor interaction, fatty acid metabolism, adhesion molecules, TGF-β signaling pathway, PPAR pathway. Further RT-qPCR might downregulate expression ASCL4 gene promoting activation PPARα, inhibiting pathway improving fibrosis In conclusion, exhibits hepatoprotective against restoring function, enhancing antioxidant capacity, its mechanism damage resistance be related improvement PPARα-TGF-β
