The emergence of drug-resistant malarial parasites has been a growing challenge to medical science safeguard public health in the malaria-endemic regions globe. With time, parasite develops newer resistance mechanisms defunct drug's action one after another. Genetic mutation is prime weapon rely upon initiate mechanism case-specific manner, following various strategies such as structural changes target protein, metabolic alterations, and tweaking drug-transported channels. In order combat these resistances, different approaches have evolved among developing inhibitors against critical enzymes pathways, combinatorial/hybrid drug therapies, exploring new targets analogues existing drugs, use resistance-reversal agents, drug-repurposing, gene blocking/altering using RNA interference CRISPR/Cas systems are prominent. However, effectiveness needs be earnestly monitored for better management disease, which demands development reliable diagnosis technique. Several methodologies investigated search suitable technique, vivo, vitro, ex vivo efficacy studies, molecular techniques. A parallel effort transform efficient method into an inexpensive portable tool rapid screening malaria masses societal landscape advocated. This review gives insight historical perspectives recent developments antimalarial discovery. Efforts made update formulated diagnose malaria. Finally, concluding remark with future perspective on subject forwarded.
