Bovine viral diarrhea virus (BVDV) is the causative agent of bovine diarrhea-mucosal disease (BVD-MD), an important in cattle that responsible for extensive economic losses to industry worldwide. Currently, several underlying mechanisms involved replication, pathogenesis, and evading host innate immunity BVDV remain be elucidated, particularly during early stage infection. To further explore BVDV-host interactions, transcriptomics proteomics profiles BVDV-infected MDBK cells were sequenced using RNA-seq iTRAQ techniques, respectively, followed by integrative analysis. Compared with mock-infected cells, a total 665 differentially expressed genes (DEGs) (391 down-regulated, 274 up-regulated) 725 proteins (DEPs) (461 264 identified. Among these, DEGs DEPs verified quantitative RT-PCR western blot. Following gene ontology (GO) annotation KEGG enrichment analysis, we determined these significantly enriched multiple cellular signaling pathways including NOD-like receptor, Toll-like TNF, NF-κB, MAPK, cAMP, lysosome, protein processing endoplasmic reticulum, lipid metabolism, apoptosis pathways. Significantly, down-regulated predominantly associated apoptosis-regulated elements, inflammatory factors, antiviral elements immunity, thus, indicating could inhibit expression facilitate replication. Meanwhile, up-regulated primarily metabolism autophagy pathways, utilize metabolic resources cell promote However, potential interactions required experimental validation. Our data provide overview changes providing basis exploring interactions.

We aimed at investigating host-virus co-metabolism during SARS-CoV-2 infection. Therefore, we extended comprehensive sex-specific, whole-body organ resolved models of human metabolism with the necessary reactions to replicate in lung as well selected peripheral organs. Using this model, obtained following key results: 1. The predicted maximal possible virus shedding rate was limited by isoleucine availability. 2. supported initial viral load depended on increase CD4+ T-cells, consistent literature. 3. During infection, changed including blood metabolome, which agreed metabolomic studies from COVID-19 patients and healthy controls. 4. could be reduced either inhibition guanylate kinase 1 or availability amino acids, e.g., diet. 5. variants differed their rates, inversely linked occurrences sequences. Taken together, study presents metabolic crosstalk between host emphasises role acid particular isoleucine. As such, it provides an example how computational modelling can complement more canonical approaches gain insight into identify potential therapeutic strategies.

P-selectin glycoprotein ligand-1 (PSGL-1) is a cell surface that binds to P-, E-, and L-selectins mediate the tethering rolling of immune cells on endothelium for migration into inflamed tissues. PSGL-1 has been identified as an interferon-γ (INF-γ)-regulated factor restricts HIV-1 infectivity, recently found possess broad-spectrum antiviral activities. Here we report expression in virus-producing impairs incorporation SARS-CoV SARS-CoV-2 spike (S) glycoproteins pseudovirions blocks pseudovirus attachment infection target cells. These findings suggest may potentially inhibit coronavirus replication PSGL-1+
