Irritable bowel syndrome (IBS) is a common disease, whose etiopathogenesis poorly understood. Human endogenous retroviruses (HERVs) originate from ancient infections of germinal cells and represent 8% our DNA. Most HERVs have become defective due to the accumulated mutations; some can, however, still be activated, their altered expressions been associated with number chronic inflammatory immune-mediated disorders, including gastrointestinal diseases. Retroviral transcription modulated by TRIM28 SETDB1, which also participate in regulation epigenetic mechanisms shaping immune system. Expressions TRIM28/SETDB1 not investigated patients affected IBS. Using PCR real-time Taqman amplification assay, we explored RNA levels HERV-H-pol, HERV-K-pol, HERV-W-pol; syncytin 1 (SYN1), SYN2, HERV-W-env; SETDB1 peripheral blood 37 IBS healthy controls (HCs) similar age. The transcript were higher than HCs for all except HERV-W-pol, significant p-values SYN1 borderline SYN2 HERV-W-env. significantly enhanced patients, while those normal range. Patients severe disease had upregulation compared mild or moderate symptoms. These findings suggest that overexpression may contribute development open way innovative therapeutic strategies.

Inflammatory bowel disease (IBD), comprising ulcerative colitis (UC) and Crohn's (CD) irritable syndrome (IBS) are distinct gastrointestinal disorders. Mycobacterium avium subspecies paratuberculosis (MAP) is implicated in IBD pathogenesis, while the roles of human endogenous retroviruses (HERVs) under investigation. We aimed (a) to investigate whether levels humoral response MAP-3865c, HERV-K envelope HERV-W against epitopes IBD/IBS patients; (b) determine frequency micronuclei patients (c) evaluate possible correlation between genomic damage response. This study investigates antibody titres MAP 3865c, env plasma from 102 IBD, 20 IBS 92 healthy controls (HCs). Micronuclei (MNi) assessed, correlating with responses patient genotype profiles. exhibited elevated those carrying GA for TNF-α showing higher anti-MAP 3865c IgG levels. A significant positive was observed MNi patients. Higher were detected both compared HCs, correlations found peptide than HCs. Our findings highlight association UC CD immune targeting HERV-Kenv. Specific genetic profiles may exacerbate inflammation, potentially amplifying patients, as indicated by frequencies.
