Abstract Licensed parenteral influenza vaccines induce systemic antibody responses and alleviate disease severity but do not efficiently prevent viral entry transmission due to the lack of local mucosal immune responses. Here, we describe intranasal booster strategy with unadjuvanted recombinant hemagglutinin (HA) following initial mRNA-LNP vaccination, Prime HA. This regimen establishes highly protective HA-specific memory in respiratory tract. Intranasal HA boosters provided significantly reduced replication compared both young old mice. Synthetic long oligodeoxynucleotides (ODNs) have found wide applications in diverse fields such as chemical biology, synthetic and genes genomes synthesis. However, traditional methods purifying ODNs present notable drawbacks, particularly inability purify ODNs, rendering ODN synthesis challenging. To address these issues, techniques coupled with non-chromatographic purification been developed. This technology makes the of feasible offers viable pathway producing extensive repeats or stable secondary structures, which are either unattainable exceedingly complex achieve using conventional methods.
