Postbiotics are functional bioactive compounds or molecules with beneficial effects on health and activities in humans livestock, produced by probiotic bacteria yeast. Several postbiotics, including enzymes, short-chain fatty acids, amino extracellular polysaccharides, microbial cell fragments, teichoic currently being widely studied. This study aimed to explore the potential of secondary metabolites Schleiferilactobacillus harbinensis LH 991 Pichia kudriavzevii B-5P as lactic acid (LAB) yeast isolated from Budu (fermented fish) which can act postbiotics through vitro rumen fermentation. The method used a completely randomized design 5 × 4, five treatments four replications. substrate diet consisted 60% forage 40% concentrate. culture mixture was 1.3 1011 CFU/mL 50%:50% ratio S. P. B-5P. inoculum concentrations this were 0% (control), 1%, 2%, 3%, 4%. Treatments arranged based differences concentration follows: T0: control (0%); T1: 1%; T2: 2%; T3: 3%; T4: T4 group showed significant increase (p < 0.01) acids (SCFA), acetate, propionate, butyrate, valerate, isobutyrate, isovalerate compared other treatments. Meanwhile, shows that there is no > effect digestibility (in dry matter digestibility, organic crude fiber digestibility). However, highly volatile total, NH3, protein synthesis. It concluded treatment 4% (T4) containing LAB increased SCFA fermentation significantly, whereas it did not affect digestibility.

Abstract Human gut is crucial for digestion, drug absorption, and overall health; however traditional in vitro animal models struggle to accurately replicate its complex mechanisms. This study introduces an innovative gut‐on‐chip based on the MIVO millifluidic device, designed faithfully human intestinal environment. CaCo‐2 HT‐29 cells were co‐cultured under different ratio dynamic flow conditions, resembling bloodstream. Intestinal tissue differentiation was assessed through Trans‐epithelial electrical resistance (TEER) measurements, Zonula Occludens‐1, Alcian blue staining. After model establishment, a second applied apical side recapitulating lumen niche. The culture conditions significantly reduced cell maturation time, obtaining differentiated layer within 7‐10 days, compared 21 days of static culture. In addition, CaCo‐2:HT‐29 co‐cultures enables finely tune mucus thicknesses barrier function, essential studying specific conditions. Furthermore, introduction double apical‐basal system recapitulated permeability characteristics more closely those observed vivo. Double‐Flow Gut‐on‐Chip described successfully validated cross‐correlate function epithelial with ratios, finally providing predictive useful development disease modelling.

Mosaicism, which is characterized by the presence of wild-type and more than one mutant allele, poses a serious problem in zygotic gene modification through clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 system. Therefore, we used pig embryos to compare editing efficiencies achieved combining electroporation lipofection using different aminopeptidase N (APN)-targeting guide RNA (gRNA) sequences. Six gRNAs (gRNA1-6) with target sequences were designed APN. Zona pellucida (ZP)-intact zygotes collected 10 h after start vitro fertilization (IVF) electroporated each gRNA their efficiency. The that lowest highest mutation rates (gRNA4 gRNA6, respectively) selected for additional assess efficiency following combined treatment.
