The interaction between colipase and lipase is known to be crucial for lipid digestion, but the roles of other factors remain unclear. This study explores dynamics pancreatic hydrolysis facilitated by colipase, with a focus on regulatory fatty acids calcium ions. Using computational modeling, including insights from AlphaFold 3.0, we elucidate structural interactions essential activity. prediction highlights significant role ions in altering lipase–colipase complex, thereby changing catalytic efficiency. We further demonstrate that slower rates are associated stronger binding affinity oleic acid, as well occupation pocket acid presence predictions provide robust framework experimental validation potential applications. These findings offer deeper into dietary digestion highlight avenues interventions addressing malnutrition.

Abstract 4,4‐Dimethylsterol (RST), rich in rice bran oil, is often considered to have good lipid‐lowering properties, and conjugated linoleic acid (CLA), a special fatty acid, also similar but the low solubility poor oxidation stability limits their application food industry. Typically, sterol esters better after esterification. However, it difficult esterify 4,4‐dimethylsterol for its additional methyl groups. Therefore, microwave‐assisted esterification of with CLA was developed this study, function 4,4‐dimethylsterol–CLA (RSE) evaluated high‐fat diet (HFD)‐induced mice model. The results showed that when reaction condition molar ratio RST 1:3 catalyzed 5% (mol%) p‐toluene sulfonic 25% dodecyl benzene maintained at microwave reactor 125 min 100°C, highest rate 93.63% acquired. Furthermore, HFD‐induced experiments demonstrated RSE could significantly reduce body weight, as well serum liver triglyceride (TG) levels, while exert hepatoprotective properties compared RST. Additionally, found affect appetite regulate expression lipid metabolism–related genes mice. These findings suggest holds potential field functional foods.
