Interfacial modular assemblies of versatile polyphenols have attracted widespread interest in surface and materials engineering. In this study, natural polyphenol (tannic acid, TA) nobiletin (NOB) can directly form binary carrier-free spherical nanoparticles (NT NPs) through synergistically driven by a variety interactions (such as hydrogen bonding, oxidative reactions, etc.). The synthesis involves polyphenolic deposition on hydrophobic NOB nanoaggregates, followed situ self-polymerization. Interestingly, the assembled NT NPs exhibit controllable dynamic changes particle size during initial stage. Ultimately, uniform appear stable, with high loading capability, enabling incorporated to preserve their function. Furthermore, vitro evaluations demonstrate that rational combination module induce apoptosis inhibit tumor metastasis for both lung cancer H1299 human fibrosarcoma HT1080 cell lines. Notably, optimized NT48 were then verified vivo experiments achieve promising synergistic anti-tumor efficacy. These findings not only provide new opportunities streamlined sensible engineering future polyphenol-based biomaterials, but also open up prospects design small-molecule nature phytochemicals.

Triple-negative breast cancer (TNBC) poses as a daunting and intricate manifestation of cancer, highlighted by few treatment options poor outlook. The crucial element in fostering tumor growth immune resistance is the polarization tumor-associated macrophages (TAMs) into M2 state within microenvironment (TME). To address this, we developed targeting peptide-chitosan-curcumin nanoparticles (M2pep-Cs-Cur NPs), targeted delivery system utilizing chitosan (Cs) carrier, curcumin (Cur) therapeutic agent, peptides for specificity. These NPs effectively inhibited TNBC cell proliferation (~ 70%) invasion 70%), while increasing responsiveness tumors to anti-PD-L1 50% survival enhancement) vitro vivo. Bioinformatics analysis suggested that Cur modulates TAM influencing key genes such COX-2, offering insights its underlying mechanisms. This study highlights potential M2pep-Cs-Cur reverse TAMs, providing promising strategy overcome immunotherapy improve outcomes.

Abstract Background Radiation exposure can cause inflammation, which etoricoxib (ET), an anti‐inflammatory drug, could potentially mitigate. Objective This study aimed to evaluate the potential effectiveness of etoricoxib‐loaded nanostructured lipid carriers (ET‐NLCs) in mitigating radiation‐induced acute lung inflammation rats. Methods Thirty‐six rats were divided into six groups. Group 1 (C): control; group 2 (ET): normal given ET (10 mg/kg) orally for 14 days; 3 (ET‐NLC): administered ET‐NLCs days. 4 (R): exposed 6 Gy whole‐body gamma radiation, untreated thereafter induce and injury. 5 (ET‐R), irradiated rats, daily (ET‐NLC‐R), Molecular, biochemical, histopathological analyses performed assess apoptosis, oxidative stress, tissue architecture. Results led a 1053% increase Bax expression 81.5% decrease Bcl‐2, indicating heightened apoptosis. treatment reversed these effects, reducing by 59.9% increasing Bcl‐2 337.4%. Additionally, reduced caspase‐3 caspase‐8 activation 54.5% 62.9%, respectively, compared radiation alone. Furthermore, demonstrated potent effects interleukin‐6 (IL‐6) tumor necrosis factor‐alpha (TNF‐α) levels 49% 39%, group. increased malondialdehyde (MDA) 388%, damage, suppressed antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GPX), superoxide dismutase (SOD). ET‐NLC decreased MDA CAT, GPX, SOD 35.7%, 4766.7%, 765.9%, restoring balance. surfactant protein (SP‐D) 4.9% control values, but restored them 14%. Histopathological analysis revealed that radiation‐exposed lungs showed thickened inter‐alveolar septa, emphysematous areas, inflammatory infiltration. exhibited only mild thickening limited cell infiltration, suggesting significant improvement Conclusions Based on results, NLCs are one most promising ways deliver drugs treating damage caused radiation.

Oleuropein is the main compound in olives, producing a relatively bitter taste for unprocessed and raw olives. It has been dramatically applied herbal traditional medicine contains several biological functions, anti‐inflammatory effects, antimicrobial characteristics, anticancer antioxidant activities. The present study dealt with cytotoxic effect, reactive oxygen species (ROS) suppressor, wound‐healing activity of oleuropein on normal skin cells. Oleuropein’s apoptotic effects were evaluated using MTT, flow cytometry, DAPI staining. Moreover, oleuropein’s possible free radical scavenging properties studied through methods, including 2, 2‐diphenyl‐1‐picrylhydrazyl (DPPH) ABTS tests. scratch assay was performed features, qRT‐PCR expression apoptosis‐associated genes. found to have effect cells at higher exposure doses. Apoptosis induced cytometry histogram treated oleuropein. results also revealed strong anti‐inflammation They suggested that more studies are necessary assess pharmacological use prevent or decrease skin‐related diseases.

Methotrexate (MTX) is a folic acid antagonist routinely used in cancer treatment, characterized by poor water solubility and low skin permeability. These issues could be mitigated using drug delivery systems, such as functionalized gold nanoparticles (AuNPs), known for their versatility unique properties. This study aimed to develop multi-shell AuNPs with MTX the improvement of antitumoral, antioxidant, biocompatibility features. Stable phosphine-coated were synthesized tailored polyethylene glycol (PEG) short-branched polyethyleneimine (PEI) moieties, followed covalent binding. Physicochemical characterization UV-vis Fourier-transform infrared spectroscopy (FTIR) spectroscopy, dynamic light scattering (DLS), scanning transmission electron microscopy (STEM), X-ray photoelectron (XPS) confirmed synthesis at each step. The antioxidant activity was determined DPPH radical scavenging assay, ferric ions' reducing power (FRAP), cupric capacity (CUPRAC) assays. Biocompatibility cytotoxicity assessed MTT LDH assays on HaCaT human keratinocytes CAL27 squamous cell carcinoma. demonstrated enhanced pronounced cytotoxic effect tumoral cells compared individual components, highlighting potential improving therapy.
