Objectives: Cyclosporine A promotes gingival fibrosis by enhancing the proliferation of fibroblasts, leading to overgrowth. The population fibroblasts is regulated cell cycle machinery, which balances growth and inhibition. Cells that detect DNA damage pause at G1/S checkpoint repair instead progressing S phase. Previous studies have linked drug-induced overgrowth response lipopolysaccharide (LPS) cyclosporine A. This research investigates effects on its mediators in LPS-treated clarify mechanisms behind cyclosporine-A-induced Methods: Semi-confluent human were treated with LPS or DMEM. Cell was evaluated counting total number cells. distribution phases analyzed using flow cytometry. Additionally, expression levels mRNAs proteins related regulators quantified reverse-transcription quantitative PCR Western blotting, respectively. Results: treatment significantly enhanced G1-S transition. It increased mRNA CDC25A CYCLIN D while decreasing those RB1, SMAD3, SMAD4. it upregulated protein CDC25A, D, CDK4, CDK6, pRB downregulated SMAD3 Conclusions: Gingival induced could be attributed these alterations.
